The haloarchaea Natrialba magadii and Haloferax volcanii, as well as the radiation-resistant bacterium Deinococcus radiodurans, were exposed to vacuum UV (VUV) radiation at the Brazilian Synchrotron Light Laboratory. Cell monolayers (containing 10(5) to 10(6) cells per sample) were prepared over polycarbonate filters and irradiated under high vacuum (10(-5) Pa) with polychromatic synchrotron radiation. N. magadii was remarkably resistant to high vacuum with a survival fraction of (3.77±0.76)×10(-2), which was larger than that of D. radiodurans (1.13±0.23)×10(-2). The survival fraction of the haloarchaea H. volcanii, of (3.60±1.80)×10(-4), was much smaller. Radiation resistance profiles were similar between the haloarchaea and D. radiodurans for fluences up to 150?J m(-2). For fluences larger than 150?J m(-2), there was a significant decrease in the survival of haloarchaea, and in particular H. volcanii did not survive. Survival for D. radiodurans was 1% after exposure to the higher VUV fluence (1350?J m(-2)), while N. magadii had a survival lower than 0.1%. Such survival fractions are discussed regarding the possibility of interplanetary transfer of viable microorganisms and the possible existence of microbial life in extraterrestrial salty environments such as the planet Mars and Jupiter's moon Europa. This is the first work to report survival of haloarchaea under simulated interplanetary conditions.  相似文献

Since the 1970s, when the Viking spacecrafts carried out experiments to detect microbial metabolism on the surface of Mars, the search for nonspecific methods to detect life in situ has been one of the goals of astrobiology. It is usually required that a methodology detect life independently from its composition or form and that the chosen biological signature point to a feature common to all living systems, such as the presence of metabolism. In this paper, we evaluate the use of microbial fuel cells (MFCs) for the detection of microbial life in situ. MFCs are electrochemical devices originally developed as power electrical sources and can be described as fuel cells in which the anode is submerged in a medium that contains microorganisms. These microorganisms, as part of their metabolic process, oxidize organic material, releasing electrons that contribute to the electric current, which is therefore proportional to metabolic and other redox processes. We show that power and current density values measured in MFCs that use microorganism cultures or soil samples in the anode are much larger than those obtained with a medium free of microorganisms or sterilized soil samples, respectively. In particular, we found that this is true for extremophiles, which have been proposed as potential inhabitants of extraterrestrial environments. Therefore, our results show that MFCs have the potential to be used for in situ detection of microbial life.  相似文献

Two types of uncertainty co-exist in the theory of evidence: discord and non-specificity. From 90s, many mathematical expressions have arisen to quantify these two parts in an evidence. An important aspect of each measure presented is the verification of a coherent set of properties. About non-specificity, so far only one measure verifies an important set of those properties. Very recently, a new measure of non-specificity based on belief intervals has been presented as an alternative measure that quantifies a similar set of properties (Yang et al., 2016). It is shown that the new measure really does not verify two of those important properties. Some errors have been found in their corresponding proofs in the original publication.  相似文献

Reproduction is a vital characteristic of life, and sex is the most common reproductive mode in the eukaryotic world. Sex and reproduction are not necessarily linked mechanisms: Sexuality without reproduction exists, while several forms of asexual reproduction are known. The occurrence of sexuality itself is paradoxical, as it is very costly in evolutionary terms. Most of the hypotheses (more than 20) attempting to explain the prevalence of sex fall into two categories: Sex either creates good gene combinations for adaptation to environments or eliminates bad gene combinations counteracting the accumulation of mutations. In spite of this apparent wealth of beneficial effects of sex, asexuality is not rare. Most eukaryotic, asexual lineages are short-lived and can only persist through the presence of sexual roots, but at least two animal groups, bdelloid rotifers and darwinulid ostracods, seem to claim the status of ancient asexuals. Research on (a)sexuality is relevant to astrobiology in a number of ways. First, strong relationships between the origin and persistence of life in extreme environments and reproductive mode are known. Second, the "habitability" of nonterrestrial environments to life greatly depends on reproductive mode. Whereas asexuals can do equally well or better in harsh environments, they fail to adapt fast enough to changing abiotic and biotic environments. Third, it has been shown that plants reproduce mainly asexually in space, and sperm production and motility in some vertebrates are hampered. Both findings indicate that extraterrestrial life under conditions different from Earth might be dominated by asexual reproduction. Finally, for exchange of biological material between planets, the choice of reproductive mode will be important.  相似文献

In the next few years, the number of catalogued exoplanets will be counted in the thousands. This will vastly expand the number of potentially habitable worlds and lead to a systematic assessment of their astrobiological potential. Here, we suggest a two-tiered classification scheme of exoplanet habitability. The first tier consists of an Earth Similarity Index (ESI), which allows worlds to be screened with regard to their similarity to Earth, the only known inhabited planet at this time. The ESI is based on data available or potentially available for most exoplanets such as mass, radius, and temperature. For the second tier of the classification scheme we propose a Planetary Habitability Index (PHI) based on the presence of a stable substrate, available energy, appropriate chemistry, and the potential for holding a liquid solvent. The PHI has been designed to minimize the biased search for life as we know it and to take into account life that might exist under more exotic conditions. As such, the PHI requires more detailed knowledge than is available for any exoplanet at this time. However, future missions such as the Terrestrial Planet Finder will collect this information and advance the PHI. Both indices are formulated in a way that enables their values to be updated as technology and our knowledge about habitable planets, moons, and life advances. Applying the proposed metrics to bodies within our Solar System for comparison reveals two planets in the Gliese 581 system, GJ 581 c and d, with an ESI comparable to that of Mars and a PHI between that of Europa and Enceladus.  相似文献

The search for unequivocal signs of life on other planetary bodies is one of the major challenges for astrobiology. The failure to detect organic molecules on the surface of Mars by measuring volatile compounds after sample heating, together with the new knowledge of martian soil chemistry, has prompted the astrobiological community to develop new methods and technologies. Based on protein microarray technology, we have designed and built a series of instruments called SOLID (for "Signs Of LIfe Detector") for automatic in situ detection and identification of substances or analytes from liquid and solid samples (soil, sediments, or powder). Here, we present the SOLID3 instrument, which is able to perform both sandwich and competitive immunoassays and consists of two separate functional units: a Sample Preparation Unit (SPU) for 10 different extractions by ultrasonication and a Sample Analysis Unit (SAU) for fluorescent immunoassays. The SAU consists of five different flow cells, with an antibody microarray in each one (2000 spots). It is also equipped with an exclusive optical package and a charge-coupled device (CCD) for fluorescent detection. We demonstrated the performance of SOLID3 in the detection of a broad range of molecular-sized compounds, which range from peptides and proteins to whole cells and spores, with sensitivities at 1-2?ppb (ng?mL?1) for biomolecules and 10? to 103 spores per milliliter. We report its application in the detection of acidophilic microorganisms in the Río Tinto Mars analogue and report the absence of substantial negative effects on the immunoassay in the presence of 50?mM perchlorate (20 times higher than that found at the Phoenix landing site). Our SOLID instrument concept is an excellent option with which to detect biomolecules because it avoids the high-temperature treatments that may destroy organic matter in the presence of martian oxidants.  相似文献

Abstract The survival strategies of one cyanobacteria colony and three terricolous lichen species from the hot subdesert of Tabernas, Spain, were studied along with topographical attributes of the area to investigate whether the protective strategies adopted by these pioneer soil colonizers are related to the environmental stressors under which they survive. A handheld Raman spectrometer was used for biomolecular characterization, while the microclimatic and topographic parameters were estimated with a Geographic Information System (GIS). We found that the survival strategies adopted by those organisms are based on different combinations of protective biomolecules, each with diverse ecophysiological functions, such as UV-radiation screening, free-energy quenching, antioxidants, and the production of different types and amounts of calcium oxalates. Our results show that the cyanobacteria community and each lichen species preferentially colonized a particular microhabitat with specific moisture and incident solar radiation levels and exhibited different adaptive mechanisms. In recent years, a number of studies have provided consistent results that suggest a link between the strategies adopted by those extremophile organisms and the microclimatic environmental parameters. To date, however, far too little attention has been paid to results from Raman analyses on dry specimens. Therefore, the results of the present study, produced with the use of our miniaturized instrument, will be of interest to future studies in astrobiology, especially due to the likely use of Raman spectroscopy at the surface of Mars. Abstract We examined a low-energy mechanism for the transfer of meteoroids between two planetary systems embedded in a star cluster using quasi-parabolic orbits of minimal energy. Using Monte Carlo simulations, we found that the exchange of meteoroids could have been significantly more efficient than previously estimated. Our study is relevant to astrobiology, as it addresses whether life on Earth could have been transferred to other planetary systems in the Solar System's birth cluster and whether life on Earth could have been transferred from beyond the Solar System. In the Solar System, the timescale over which solid material was delivered to the region from where it could be transferred via this mechanism likely extended to several hundred million years (as indicated by the 3.8-4.0?Ga epoch of the Late Heavy Bombardment). This timescale could have overlapped with the lifetime of the Solar birth cluster (~100-500?Myr). Therefore, we conclude that lithopanspermia is an open possibility if life had an early start. Adopting parameters from the minimum mass solar nebula, considering a range of planetesimal size distributions derived from observations of asteroids and Kuiper Belt objects and theoretical coagulation models, and taking into account Oort Cloud formation models, we discerned that the expected number of bodies with mass>10?kg that could have been transferred between the Sun and its nearest cluster neighbor could be of the order of 10(14) to 3·10(16), with transfer timescales of tens of millions of years. We estimate that of the order of 3·10(8)·l (km) could potentially be life-bearing, where l is the depth of Earth's crust in kilometers that was ejected as the result of the early bombardment.
