The Atacama Desert has long been considered a good Mars analogue for testing instrumentation for planetary exploration, but very few data (if any) have been reported about the geomicrobiology of its salt-rich subsurface. We performed a Mars analogue drilling campaign next to the Salar Grande (Atacama, Chile) in July 2009, and several cores and powder samples from up to 5?m deep were analyzed in situ with LDChip300 (a Life Detector Chip containing 300 antibodies). Here, we show the discovery of a hypersaline subsurface microbial habitat associated with halite-, nitrate-, and perchlorate-containing salts at 2?m deep. LDChip300 detected bacteria, archaea, and other biological material (DNA, exopolysaccharides, some peptides) from the analysis of less than 0.5?g of ground core sample. The results were supported by oligonucleotide microarray hybridization in the field and finally confirmed by molecular phylogenetic analysis and direct visualization of microbial cells bound to halite crystals in the laboratory. Geochemical analyses revealed a habitat with abundant hygroscopic salts like halite (up to 260?g kg(-1)) and perchlorate (41.13?μg g(-1) maximum), which allow deliquescence events at low relative humidity. Thin liquid water films would permit microbes to proliferate by using detected organic acids like acetate (19.14?μg g(-1)) or formate (76.06?μg g(-1)) as electron donors, and sulfate (15875?μg g(-1)), nitrate (13490?μg g(-1)), or perchlorate as acceptors. Our results correlate with the discovery of similar hygroscopic salts and possible deliquescence processes on Mars, and open new search strategies for subsurface martian biota. The performance demonstrated by our LDChip300 validates this technology for planetary exploration, particularly for the search for life on Mars.  相似文献

Antibody microarrays are becoming frequently used tools for analytical purposes. A key factor for optimal performance is the stability of the immobilized (capturing) antibodies as well as those that have been fluorescently labeled to achieve the immunological test (tracers). This is especially critical for long-distance transport, field testing, or planetary exploration. A number of different environmental stresses may affect the antibody integrity, such as dryness, sudden temperature shift cycles, or, as in the case of space science, exposure to large quantities of the highly penetrating gamma radiation. Here, we report on the effect of certain stabilizing solutions for long-term storage of printed antibody microarrays under different conditions. We tested the effect of gamma radiation on printed and freeze- or vacuum-dried fluorescent antibodies at working concentrations (tracer antibodies), as well as the effect of multiple cycles of sudden and prolonged temperature shifts on the stability of fluorescently labeled tracer antibody cocktails. Our results show that (i) antibody microarrays are stable at room temperature when printed on stabilizing spotting solutions for at least 6 months, (ii) lyophilized and vacuum-dried fluorescently labeled tracer antibodies are stable for more than 9 months of sudden temperature shift cycles (-20°C to 25°C and 50°C), and (iii) both printed and freeze- or vacuum-dried fluorescent tracer antibodies are stable after several-fold excess of the dose of gamma radiation expected during a mission to Mars. Although different antibodies may exhibit different susceptibilities, we conclude that, in general, antibodies are suitable for use in planetary exploration purposes if they are properly treated and stored with the use of stabilizing substances.  相似文献

We review the current knowledge and understanding of dust in the inner solar system. The major sources of the dust population in the inner solar system are comets and asteroids, but the relative contributions of these sources are not quantified. The production processes inward from 1 AU are: Poynting-Robertson deceleration of particles outside of 1 AU, fragmentation into dust due to particle-particle collisions, and direct dust production from comets. The loss processes are: dust collisional fragmentation, sublimation, radiation pressure acceleration, sputtering, and rotational bursting. These loss processes as well as dust surface processes release dust compounds in the ambient interplanetary medium. Between 1 and 0.1 AU the dust number densities and fluxes can be described by inward extrapolation of 1 AU measurements, assuming radial dependences that describe particles in close to circular orbits. Observations have confirmed the general accuracy of these assumptions for regions within 30° latitude of the ecliptic plane. The dust densities are considerably lower above the solar poles but Lorentz forces can lift particles of sizes < 5 μm to high latitudes and produce a random distribution of small grains that varies with the solar magnetic field. Also long-period comets are a source of out-of-ecliptic particles. Under present conditions no prominent dust ring exists near the Sun. We discuss the recent observations of sungrazing comets. Future in-situ experiments should measure the complex dynamics of small dust particles, identify the contribution of cometary dust to the inner-solar-system dust cloud, and determine dust interactions in the ambient interplanetary medium. The combination of in-situ dust measurements with particle and field measurements is recommended.  相似文献

The Ulysses spacecraft has been orbiting the Sun on a highly inclined ellipse almost perpendicular to the ecliptic plane (inclination 79°, perihelion distance 1.3 AU, aphelion distance 5.4 AU) since it encountered Jupiter in 1992. The in situ dust detector on board continuously measured interstellar dust grains with masses up to 10−13 kg, penetrating deep into the solar system. The flow direction is close to the mean apex of the Sun’s motion through the solar system and the grains act as tracers of the physical conditions in the local interstellar cloud (LIC). While Ulysses monitored the interstellar dust stream at high ecliptic latitudes between 3 and 5 AU, interstellar impactors were also measured with the in situ dust detectors on board Cassini, Galileo and Helios, covering a heliocentric distance range between 0.3 and 3 AU in the ecliptic plane. The interstellar dust stream in the inner solar system is altered by the solar radiation pressure force, gravitational focussing and interaction of charged grains with the time varying interplanetary magnetic field. We review the results from in situ interstellar dust measurements in the solar system and present Ulysses’ latest interstellar dust data. These data indicate a 30° shift in the impact direction of interstellar grains w.r.t. the interstellar helium flow direction, the reason of which is presently unknown.  相似文献

The ESA mission Rosetta, launched on March 2nd, 2004, carries an instrument suite to the comet 67P/Churyumov-Gerasimenko. The COmetary Secondary Ion Mass Anaylzer – COSIMA – is one of three cometary dust analyzing instruments onboard Rosetta. COSIMA is based on the analytic measurement method of secondary ion mass spectrometry (SIMS). The experiment’s goal is in-situ analysis of the elemental composition (and isotopic composition of key elements) of cometary grains. The chemical characterization will include the main organic components, present homologous and functional groups, as well as the mineralogical and petrographical classification of the inorganic phases. All this analysis is closely related to the chemistry and history of the early solar system. COSIMA covers a mass range from 1 to 3500 amu with a mass resolution mm @ 50% of 2000 at mass 100 amu. Cometary dust is collected on special, metal covered, targets, which are handled by a target manipulation unit. Once exposed to the cometary dust environment, the collected dust grains are located on the target by a microscopic camera. A pulsed primary indium ion beam (among other entities) releases secondary ions from the dust grains. These ions, either positive or negative, are selected and accelerated by electrical fields and travel a well-defined distance through a drift tube and an ion reflector. A microsphere plate with dedicated amplifier is used to detect the ions. The arrival times of the ions are digitized, and the mass spectra of the secondary ions are calculated from these time-of-flight spectra. Through the instrument commissioning, COSIMA took the very first SIMS spectra of the targets in space. COSIMA will be the first instrument applying the SIMS technique in-situ to cometary grain analysis as Rosetta approaches the comet 67P/Churyumov-Gerasimenko, after a long journey of 10 years, in 2014.  相似文献

A cosmic dust detector is currently being developed using a piezoelectric lead zirconate titanate (PZT) element. The characteristics of the multilayered detector (MD), which was composed of one hundred PZT disks, were investigated by bombarding it with hypervelocity iron particles supplied by a Van de Graaff accelerator. It was confirmed that there was a linear relationship between the signal amplitude observed from MD and the momentum of the particles. As compared with the single-layered detector (SD) that was composed of one PZT disk, it was found that the sensitivity of MD was ∼3 times higher than that of SD within the limits of the experimental conditions.  相似文献

The MSTIDs are wave-like perturbations of the ionospheric plasma, which cause the most common ionospheric disturbances in mid-latitude regions. Generally the MSTIDs have velocities of several hundred meters per second and wavelengths of several hundred kilometers. The wave-like effect of the MSTID is one of the main obstacles for accurate interpolation of ionospheric corrections in a medium-scale reference GPS network. In this paper we show a new method of detecting and modeling MSTIDs using dense German GPS network. The between-epoch single difference ionospheric delays from a medium scale dense GPS network are used to estimate the parameter of the MSTID e.g. amplitude, wavelength and velocity. The efficiency of the approach is tested with data from about 320 GPS stations in and near Germany. A MSTID wave moving from east to west across Germany was observed at September 27 in 2009. Its wavelength is about 302 km, with a period of ∼7 min and velocity of about 700 m/s.  相似文献

Quiescent prominences can be modeled as thin slabs of cold, dense plasma embedded in the much hotter and rarer solar corona. Although their global shape is rather irregular, they are often characterised by an internal structure consisting of a large number of thin, parallel threads piled together. Prominences often display periodic disturbances mostly observed in the Doppler displacement of spectral lines and with an amplitude typically of the order of or smaller than 2–3 km?s?1, a value which seems to be much smaller than the characteristic speeds of the prominence plasma (namely the Alfvén and sound velocities). Two particular features of these small amplitude prominence oscillations are that they seem to damp in a few periods and that they seem not to affect the whole prominence structure. In addition, in high spatial resolution observations, in which threads can be discerned, small amplitude oscillations appear to be clearly associated to these fine structure constituents. Prominence seismology tries to bring together the results from these observations (e.g. periods, wavelengths, damping times) and their theoretical modeling (by means of the magnetohydrodynamic theory) to gain insight into physical properties of prominences that cannot be derived from direct observation. In this paper we discuss works that have not been described in previous reviews, namely the first seismological application to solar prominences and theoretical advances on the attenuation of prominence oscillations.  相似文献

The detection of organic molecules of unambiguous biological origin is fundamental for the confirmation of present or past life. Planetary exploration requires the development of miniaturized apparatus for in situ life detection. Analytical techniques based on mass spectrometry have been traditionally used in space science. Following the Viking landers, gas chromatography-mass spectrometry (GC-MS) for organic detection has gained general acceptance and has been used successfully in the Cassini–Huygens mission to Titan. Microfluidics allows the development of miniaturized capillary electrophoresis devices for the detection of important molecules for life, like amino acids or nucleobases. Recently, a new approach is gaining acceptance in the space science community: the application of the well-known, highly specific, antibody–antigen affinity interaction for the detection and identification of organics and biochemical compounds. Antibodies can specifically bind a plethora of structurally different compounds of a broad range of molecular sizes, from amino acids level to whole cells. Antibody microarray technology allows us to look for the presence of thousands of different compounds in a single assay and in just one square centimeter. Herein, we discuss several important issues—most of which are common with other instruments dealing with life signature detection in the solar system—that must be addressed in order to use antibody microarrays for life detection and planetary exploration. These issues include (1) preservation of biomarkers, (2) the extraction techniques for biomarkers, (3) terrestrial analogues, (4) the antibody stability under space environments, (5) the selection of unequivocal biomarkers for the antibody production, or (6) the instrument design and implementation.  相似文献
