Doppler ultrasound is frequently used for monitoring circulating microbubbles during decompression to assess the symptoms of Decompression Sickness (DCS). This analysis was carried out to evaluate its effectiveness for screening symptoms of DCS during simulated extravehicular activities (EVA). The information from various hypobaric chamber studies carried out at the NASA Johnson Space Center, Houston, TX was used in this analysis (n = 516). The circulating microbubbles were detected in the precordial area in 42% (218/516), and symptoms were reported in 16% (81/516) of these exposures. The accuracy of Doppler-detectable bubbles (Spencer grades) on all symptoms of DCS was examined by calculating measures of sensitivity and specificity. The efficacy of Doppler as a screening device was examined by calculating their positive predictive value (PPV) and negative predictive value (NPV). The results of these analyses indicated that the sensitivity of Doppler decreased, and the PPV increased with higher Spencer grades. However, the likelihood of detecting true negative cases (NPV) was consistently higher with all bubble grades. Due to the high false-positive rate and low prior probabilities of the risk of DCS, Doppler was found to be more useful to identify those who did not develop DCS, than to detect positive cases of DCS in the simulated EVA exposures.  相似文献

Formation and subsequent evolution of gas bubbles in blood and tissues of subjects exposed to decompression are casual processes in their nature. Such character of bubbling processes in a body predetermines probabilistic character of decompression sickness (DCS) incidence in divers, aviators and astronauts. Our original probabilistic theory of decompression safety is based on stochastic models of these processes and on the concept of critical volume of a free gas phase in body tissues. From positions of this theory, the probability of DCS incidence during single-stage decompressions and during hypobaric decompressions under EVA in particular, is defined by the distribution of possible values of nucleation efficiency in "pain" tissues and by its critical significance depended on the parameters of a concrete decompression. In the present study the following is shown: 1) the dimensionless index of critical nucleation efficiency for "pain" body tissues is a more adequate index of decompression stress in comparison with Tissue Ratio, TR; 2) a priory the decompression under EVA performed according to the Russian protocol is more safe than decompression under EVA performed in accordance with the U.S. protocol; 3) the Russian space suit operated at a higher pressure and having a higher "rigidity" induces a stronger inhibition of mechanisms of cavitation and gas bubbles formation in tissues of a subject located in it, and by that provides a more considerable reduction of the DCS risk during real EVA performance.  相似文献

A 10.2 psi staged-decompression schedule or a 4-hour preoxygenation at 14.7 psi is required prior to extravehicular activity (EVA) to reduce decompression sickness (DCS) risk. Results of recent research at the Air Force Research Laboratory (AFRL) showed that a 1-hour resting preoxygenation followed by a 4-hour, 4.3 psi exposure resulted in 77% DCS risk (N=26), while the same profile beginning with 10 min of exercise at 75% of VO2peak during preoxygenation reduced the DCS risk to 42% (P<.03; N=26). A 4-hour preoxygenation without exercise followed by the 4.3 psi exposure resulted in 47% DCS risk (N=30). The 1-hour preoxygenation with exercise and the 4-hour preoxygenation without exercise results were not significantly different. Elimination of either 3 hours of preoxygenation or 12 hours of staged-decompression are compelling reasons to consider incorporation of exercise-enhanced preoxygenation.  相似文献

During extravehicular activities (EVA) outside the spacecraft, astronauts have to work under reduced pressure in a space suit. This pressure reduction induces the risk of decompression sickness (DCS) by the formation of gas bubbles from excess nitrogen dissolved in the organism by breathing air at normal pressure. Under laboratory conditions the gas bubbles moving in the blood stream can be detected by the non-invasive ultrasonic Doppler method. By early detection of excessive bubble formation the development of DCS symptoms may be prevented by early application of preventative measures. The method could also be useful when applied in the space suit in order to compare the results of laboratory tests with operational results, because there is a discrepancy according to the DCS risk of laboratory experiments and actual EVA missions, where no symptoms have been reported yet. A prototype Doppler sensor has been developed and implemented in the Russian Orlan suit. To investigate the feasibility of this method under simulated space conditions, the equipment has been used in a series of 12 thermovacuum chamber tests with suited subjects, where intravenous bubble formation was compared to unsuited control experiments. In more than 50% of the suited tests good Doppler recordings could be achieved. In some cases with unsatisfying results the signal could be improved by breathholding. Although the results do not yet allow any conclusion about a possible difference between suited and unsuited subjects due to the small number of tests performed, the method proved its feasibility for use in EVA suits and should be further developed to enhance the safety of EVA procedures.  相似文献

The aim of this paper is to review the main results of medical support of 78 two-person extravehicular activities (EVAs) which have been conducted in the Mir Space Program. Thirty-six male crewmembers participated in these EVAs. Maximum length of a space walk was equal to 7 h 14 min. The total duration of all space walks reached 717.1 man-hours. The maximum frequency of EVA's execution was 10 per year. Most of the EVAs (67) have been performed at mission elapsed time ranging from 31 to 180 days. The oxygen atmosphere of the Orlan space suit with a pressure of 40 kPa in combination with the normobaric cabin environment and a short (30 min) oxygen prebreathe protocol have minimized the risk of decompression sickness (DCS). There has been no incidence of DCS during performed EVAs. At the peak activity, metabolic rates and heart rates increased up to 9.9-13 kcal/min and 150-174 min-1, respectively. The medical problems have centred on feeling of moderate overcooling during a rest period in a shadow after the high physical loads, episodes with tachycardia accompanied by cardiac rhythm disorders at the moments of emotional stress, pains in the muscles and general fatigue after the end of a hard EVA. The increase of nonesterified fatty acid (NEFA) concentration in plasma was observed in rats subjected to hypokinesia for 1-60 days. In the period of recovery (7 and 21 days after 60 days immobilization) the content of NEFA returned to control values. The increase of fatty acid release from adipose tissue was observed in hypokinetic rats, however the stimulation of lipolysis by norepinephrine was lower in rats exposed to hypokinesis. The decrease of the binding capacity and a diminished number of beta-adrenergic receptors were found in animals after hypokinesia. The augmentation of the incorporation of glucose into lipids and the marked increase in the stimulation of lipogenesis by insulin were found in adipose tissue of rats subjected to long-term hypokinesia. These results showed an important effect of hypokinesia on lipid mobilization, on lipogenesis and on the processes of hormone regulation in adipose tissue.  相似文献

The ultrasonic bubble detection technique has been used as a versatile tool in the investigation of hypobaric decompression procedures for the last two decades. However, this method can only detect relatively large bubbles > 30-50 micrometers (usually in venous blood) and not the very first stages of bubble formation. Recently, the mechanical oscillator technique has been applied in air diving trials in order to detect small blood density changes due to hypothetical fluid shifts. However, sharp density drops were found in the decompression process, and these have been attributed to gas bubble formation. In this study, this method was applied together with Doppler controls to a series of altitude decompressions to 300 and 500 hPa, which represents the range of EVA operational pressures currently used or planned. Six subjects participated in both series; the oscillator sample tube could be pressurized. By the application of pressure on a blood sample containing questionable gas bubbles, these are reduced in volume, like in recompression therapy, and thus the density of the sample is increased. This effect has been shown in all 6 subjects undergoing decompression to 300 hPa, whereas in the 500 hPa series only 3 out of 6 subjects corresponded with gas phase formation immediately after decompression. Typically, Doppler bubbles appeared only after some latency time between 20 and 60 min; after this time, the density effect had nearly vanished. It is concluded that the mechanical oscillator technique represents a complementary research instrument to the Doppler method, because it is able to deliver a quantitative resolution of the early stage of gas phase formation.  相似文献

Architectural changes in trabecular bone by osteoporosis were utilized as a model for the changes which probably occur in human bone while exposed to microgravity conditions. Although there are many concerns about microgravity-induced bone loss, little is known about the impact of microgravity on the three-dimensional architecture of the skeleton. 50 (level L3) and 57 (level L4) vertebral bones harvested from human cadavers were investigated by computed tomography (CT) and quantified in terms of bone mineral density (BMD). Based on the symbol-encoded transformed CT-images, five measures of complexity were developed which quantify the structural composition of the trabecular bone. This quantification determines the bone architecture as a whole. Depending on the specific measure of complexity and its relation to BMD, a 5-10% change of BMD is related to a 5-90% change in structural composition. The method requires a non-invasive CT-procedure of the lumbar spine resulting in a radiation exposure of about 30 microSv effective dose. The technique is useful for the evaluation of the bone status of space-flying, personnel as well as for patients on ground. Grant numbers: BMH1-CT92-0296.  相似文献

Lipid biomarkers are widely used to study the earliest life on Earth and have been invoked as potential astrobiological markers, but few studies have assessed their survival and persistence in geothermal settings. Here, we investigate lipid preservation in active and inactive geothermal silica sinters, with ages of up to 900 years, from Champagne Pool, Waiotapu, New Zealand. Analyses revealed a wide range of bacterial biomarkers, including free and bound fatty acids, 1,2-di-O-alkylglycerols (diethers), and various hopanoids. Dominant archaeal lipids include archaeol and glycerol dialkyl glycerol tetraethers (GDGTs). The predominance of generally similar biomarker groups in all sinters suggests a stable microbial community throughout Champagne Pool's history and indicates that incorporated lipids can be well preserved. Moreover, subtle differences in lipid distributions suggest that past changes in environmental conditions can be elucidated. In this case, higher archaeol abundances relative to the bacterial diethers, a greater proportion of cyclic GDGTs, the high average chain length of the bacterial diethers, and greater concentrations of hopanoic acids in the older sinters all suggest hotter conditions at Champagne Pool in the past.  相似文献

The quantitative changes in nucleic acids and chromatin breakdown were followed in blood, thymus and spleen in rats after 14 day flights on board the biosatellites Cosmos-1887 and Cosmos-2044. Quantitative nucleic acid changes within 8-11 h after landing were only mild, most statistically non-significant. An analysis at 48 h after landing showed a marked decrease in a total content of DNA and RNA in spleen and thymus. Within 8-11 h after landing, the symptoms of chromatin breakdown were found as is seen in an increased concentration of its fragments-polydeoxyribonucleotides. The obtained results show that a partial adaptation to microgravity occurs up to flight day 14 in lymphoid organs. Adaptation is accompanied with a reappearing of the sensitive cells. Their chromatin breaks down, then, in a final phase of flight due to hypergravity stress manifesting itself by a temporary increase in polydeoxyribonucleotide concentration several hours after landing. The results are discussed in relation to the changes in chosen parameters after shorter or more prolonged flights.  相似文献

Gravitational unloading results in pronounced atrophy of m.soleus. Probably, the output of NO is controlled by the muscle activity. We hypothesized that NO may be involved in the protein metabolism and increase of its concentration in muscle can prevent atrophic changes induced by gravitational unloading. In order to test the hypothesis we applied NO donor l-arginine during gravitational unloading. 2.5-month-old male Wistar rats weighing 220–230g were divided into sedentary control group (CTR, n=7), 14-day hindlimb suspension (HS, n=7), 14 days of hindlimb suspension+l-arginine (HSL, n=7) (with a daily supplementation of 500 mg/kg wt l-arginine) and 14 days of hindlimb suspension+l-NAME (HSN, n=7) (90 mg/kg wt during 14 days). Cross sectional area (CSA) of slow twitch (ST) and fast twitch (FT) soleus muscle fibers decreased by 45% and 28% in the HS group (p<0.05) and 40% and 25% in the HSN group, as compared to the CTR group (p<0.05), respectively. CSA of ST and FT muscle fibers were 25% and 16% larger in the HSL group in comparison with the HS group (p<0.05), respectively. The atrophy of FT muscle fibers in the HSL group was completely prevented since FT fiber CSA had no significant differences from the CTR group. In HS group, the percentage of fibers revealing either gaps/disruption of the dystrophin layer of the myofiber surface membrane increased by 27% and 17%, respectively, as compared to the controls (CTR group, p<0.05). The destructions in dystrophin layer integrity and reductions of desmin content were significantly prevented in HSL group. NO concentration decreased by 60% in the HS group (as well as HSN group) and at the same time no changes were detectable in the HSL group. This fact indicates the compensation of NO content in the unloaded muscle under l-arginine administration. The levels of atrogin-1 mRNA were considerably altered in suspended animals (HS group: plus 27%, HSL group: minus 13%) as compared to the control level. Conclusion: l-arginine administration allows maintaining NO concentration in m.soleus at the level of cage control group, prevents from dystrophin layer destruction, decreases the atrogin mRNA concentration in the muscle and atrophy level under gravitational unloading.  相似文献

Bed rest (BR) induces significant urinary and blood electrolyte changes, but little is known about the effect of fluid and salt supplements (FSS) on catabolism, hydration and electrolytes. The aim was to measure the effect of FSS on catabolism, body hydration and electrolytes during BR.

Studies were done during 7 days of a pre-bed rest period and during 30 days of a rigorous bed rest period. Thirty male athletes aged, 24.6±7.6 years were chosen as subjects. They were divided into three groups: unsupplemented ambulatory control subjects (UACS), unsupplemented bed rested subjects (UBRS) and supplemented bed rested subjects (SBRS). The UBRS and SBRS groups were kept under a rigorous bed rest regime for 30 days. The SBRS daily took 30 ml water per kg body weight and 0.1 sodium chloride per kg body weight.

Plasma sodium (Na), potassium (K), calcium (Ca) and magnesium (Mg) levels, urinary Na, K, Ca and Mg excretion, plasma osmolality, plasma protein level, whole blood hemoglobin (Hb) and hematocrit (Hct) level increased significantly (p≤0.05), while plasma volume (PV), body weight, body fat, peak oxygen uptake, food and fluid intake decreased significantly (p≤0.05) in the UBRS group when compared with the SBRS and UACS groups. In contrast, plasma and urinary electrolytes, osmolality, protein level, whole blood Hct and Hb level decreased significantly (p≤0.05), while PV, fluid intake, body weight and peak oxygen uptake increased significantly (p≤0.05) in the SBRS group when compared with the UBRS group. The measured parameters did not change significantly in the UACS group when compared with their baseline control values.

The data indicate that FSS stabilizes electrolytes and body hydration during BR, while BR alone induces significant changes in electrolytes and body hydration. We conclude that FSS may be used to prevent catabolism and normalize body hydration status and electrolyte values during BR.  相似文献

Suppression of the immune system after space flights of different duration has been reported earlier by Konstantinova [Immune system in extreme conditions, Space immunology. B. 59. M. Science 1988. 289p. (in Russian) [4]; Immunoresistance of man in space flight, Acta Astronautica 23 (1991) 123–127 [5]]. Changes in T- and B-mediated activities of the immune system were demonstrated during and after space flight. However, the influence of the space flight conditions on the allergic status of cosmonauts and astronauts is still unclear. The goal of this investigation was to analyze total blood IgE levels, specific IgE-antibodies and interleukin-4 in blood of Russian crew members before and after space flights to the International Space Station (ISS) and during a long-term isolation study. For this purpose, we used the ELISA assays as well as other special kits. It was noticed that four out of nine cosmonauts had high total serum IgE (more than normal clinical values of 120 IU/ml). At the same time, there were no statistically significant changes in serum IgE levels before and after long-term space flights (128–195 days). A similar situation was observed regarding preflight IgE levels of cosmonauts who performed short-term flights (7–11 days). However, seven out of 11 cosmonauts had increased IgE level in blood post short flights as compared with pre-flight values. We also measured specific IgE-antibodies, because their high concentration may cause the increased production of total IgE indicating sensitization of cosmonauts. This becomes more important when humans spend a longer time in the closed environment of a space vehicle. Also our ground-based investigations showed that a stay in such conditions does not enhance sensitization to allergens (total number of tested allergens 27) including food, inhalants and cross-reactive proteins. Serum interleukin-4 level measured after short- and long-term space flights was identical. A linear correlation between levels of immunoglobulin E and interleukin-4 also was not significant. Despite the fact that our investigations did not establish any influence of space flight on sensitization and development of immediate-type allergic reactions, they demonstrated the necessity to control the allergic status of cosmonauts very carefully both before and after space flights. At the same time, it is necessary to pay special attention to outcomes of atopic individuals with high pre-flight level of total blood IgE.  相似文献

Extremely acid (pH <1) saline lakes and groundwaters existed in the mid-Permian of the mid-continent of North America. Modern counterparts have been found in acid saline lake systems throughout southern Australia. We compare and contrast the Permian Opeche Shale of North Dakota and Nippewalla Group of Kansas to modern Australian salt lakes in southern Western Australia and in northwest Victoria. With the exception of some minor variations in pH, evaporite mineralogy, and water geochemistry, the Permian and modern systems are similar and characterized by: (1) ephemeral saline continental playas hosted by red siliciclastic sediments, (2) evaporite minerals, including abundant sulfates, (3) Al-Fe-Si-rich waters with low pH values, (4) acidophilic microbes, and (5) paucity of carbonates. The composition of these terrestrial systems is strikingly similar to compositional data returned from the martian surface. Specifically, both Earth and martian systems have high amounts of iron oxides and sulfates, and little, if any, carbonates. We propose that the modern and ancient terrestrial acid saline environments may be good analogs for possible environments on Mars.  相似文献

We measured the urine amino acid distribution patterns before, during and after space flight on the Space Shuttle. The urine samples were collected on two separate flights of the space shuttle. The first flight lasted 9.5 days and the second flight 15 days. Urine was collected continuously on 8 subjects for the period beginning 10 d before launch to 6 d after landing. Results: In contrast to the earlier Skylab missions where a pronounced amino aciduria was found, on shuttle the urinary amino acids showed little change with spaceflight except for a marked decrease in all of the amino acids on FD (flight day) 1 (p<0.05) and a reduction in isoleucine and valine on FD3 and FD4 (p<0.05). Conclusions: (i) Amino aciduria is not an inevitable consequence of space flight. (ii) The occurrence of amino aciduria, like muscle protein breakdown is a mission specific effect rather than part of the general human response to microgravity.  相似文献

We report on the use of a portable instrument for microbial detection in the Mojave Desert soil and the potential for its use on Mars. The instrument is based on native fluorescence and employs four excitation wavelengths combined with four emission wavelengths. A soil dilution series in which known numbers of Bacillus subtilis spores were added to soil was used to determine the sensitivity of the instrument. We found that the fluorescence of the biological and organic components of the desert soil samples studied can be as strong as the fluorescence of the mineral component of these soils. Using the calibration derived from B. subtilis spores, we estimated that microbial content at our primary sampling site was 10(7) bacteria per gram of soil, a level confirmed by phospholipid fatty acid analysis. At a nearby site, but in a slightly different geological setting, we tested the instrument's ability to map out microbial concentrations in situ. Over a ~50 m diameter circle, soil microbial concentrations determined with the B. subtilis calibration indicate that the concentrations of microorganisms detected varies from 10(4) to 10(7) cells per gram of soil. We conclude that fluorescence is a promising method for detecting soil microbes in noncontact applications in extreme environments on Earth and may have applications on future missions to Mars.  相似文献

This paper gives a summary of the principal hematological and immunological changes observed in crews after space flight. Reduction of red blood cell mass (2-21%) and of hemoglobin mass (12-33%) is generally observed after the US and Soviet space missions. The changes are accompanied with a loss of plasma volume (4-16%). Erythrocyte and hemoglobin concentrations in the blood remain constant, suggesting that the changes are driven by a feed-back mechanism. Immunological changes consist mainly of reduced T-lymphocyte reactivity. The results of the 96-day and 140-day Salyut-6 missions suggest that the adaptation of the immune system to spaceflight occurs in two stages: the first takes place during the first 2-3 months in space, the second follows and consists of further weakening of the immune response. Our experiments with human lymphocytes in vitro indicate that high-g enhance, whereas low-g depress lymphocyte activity. Finally, our investigations to be performed on Spacelab are described.  相似文献

Abstract Microbial mats are self-sustained, functionally complex ecosystems that make good models for the understanding of past and present microbial ecosystems as well as putative extraterrestrial ecosystems. Ecological theory suggests that the composition of these communities might be affected by nutrient availability and disturbance frequency. We characterized two microbial mats from two contrasting environments in the oligotrophic Cuatro Ciénegas Basin: a permanent green pool and a red desiccation pond. We analyzed their taxonomic structure and composition by means of 16S rRNA clone libraries and metagenomics and inferred their metabolic role by the analysis of functional traits in the most abundant organisms. Both mats showed a high diversity with metabolically diverse members and strongly differed in structure and composition. The green mat had a higher species richness and evenness than the red mat, which was dominated by a lineage of Pseudomonas. Autotrophs were abundant in the green mat, and heterotrophs were abundant in the red mat. When comparing with other mats and stromatolites, we found that taxonomic composition was not shared at species level but at order level, which suggests environmental filtering for phylogenetically conserved functional traits with random selection of particular organisms. The highest diversity and composition similarity was observed among systems from stable environments, which suggests that disturbance regimes might affect diversity more strongly than nutrient availability, since oligotrophy does not appear to prevent the establishment of complex and diverse microbial mat communities. Hydrothermal pyrolysis experiments were performed to assess condensation (dehydration) reactions to amide, ester, and nitrile functionalities from lipid precursors. Beside product formation, organic compound alteration and stability were also evaluated. Mixtures of nonadecanoic acid, hexadecanedioic acid, or hexadecanamide with water, ammonium bicarbonate, and oxalic acid were heated at 300 degrees C for 72 h. In addition, mixtures of ammonium bicarbonate and oxalic acid solutions were used to test the abiotic formation of organic nitrogen compounds at the same temperature. The resulting products were condensation compounds such as amides, nitriles, and minor quantities of N-methylalkyl amides, alkanols, and esters. Mixtures of alkyl amide in water or oxalic acid yielded mainly hydrolysis and dehydration products, and with ammonium bicarbonate and oxalic acid the yield of condensation products was enhanced. The synthesis experiments with oxalic acid and ammonium bicarbonate solutions yielded homologous series of alkyl amides, alkyl amines, alkanes, and alkanoic acids, all with no carbon number predominances. These organic nitrogen compounds are stable and survive under the elevated temperatures of hydrothermal fluids.  相似文献
