The origin and early evolution of membrane channels   总被引:3,自引:0,他引:3
The origin and early evolution of ion channels are considered from the point of view that the transmembrane segments of membrane proteins are structurally quite simple and do not require specific sequences to fold. We argue that the transport of solute species, especially ions, required an early evolution of efficient transport mechanisms, and that the emergence of simple ion channels was protobiologically plausible. We also argue that, despite their simple structure, such channels could possess properties that, at the first sight, appear to require markedly greater complexity. These properties can be subtly modulated by local modifications to the sequence rather than global changes in molecular architecture. In order to address the evolution and development of ion channels, we focus on identifying those protein domains that are commonly associated with ion channel proteins and are conserved throughout the three main domains of life (Eukarya, Bacteria, and Archaea). We discuss the potassium-sodium-calcium superfamily of voltage-gated ion channels, mechanosensitive channels, porins, and ABC-transporters and argue that these families of membrane channels have sufficiently universal architectures that they can readily adapt to the diverse functional demands arising during evolution.  相似文献

Morphological, compositional, and biological evidence indicates the presence of numerous well-developed microbial hyphae structures distributed within four different sample splits of the Nakhla meteorite obtained from the British Museum (allocation BM1913,25). By examining depth profiles of the sample splits over time, morphological changes displayed by the structures were documented, as well as changes in their distribution on the samples, observations that indicate growth, decay, and reproduction of individual microorganisms. Biological staining with DNA-specific molecular dyes followed by epifluorescence microscopy showed that the hyphae structures contain DNA. Our observations demonstrate the potential of microbial interaction with extraterrestrial materials, emphasize the need for rapid investigation of Mars return samples as well as any other returned or impactor-delivered extraterrestrial materials, and suggest the identification of appropriate storage conditions that should be followed immediately after samples retrieved from the field are received by a handling/curation facility. The observations are further relevant in planetary protection considerations as they demonstrate that microorganisms may endure and reproduce in extraterrestrial materials over long (at least 4 years) time spans. The combination of microscopy images coupled with compositional and molecular staining techniques is proposed as a valid method for detection of life forms in martian materials as a first-order assessment. Time-resolved in situ observations further allow observation of possible (bio)dynamics within the system.  相似文献

Bacterial spores have been considered as microbial life that could survive interplanetary transport by natural impact processes or human spaceflight activity. Deposition of terrestrial microbes or their biosignature molecules onto the surface of Mars could negatively impact life detection experiments and planetary protection measures. Simulated Mars solar radiation, particularly the ultraviolet component, has been shown to reduce spore viability, but its effect on spore germination and resulting production of biosignature molecules has not been explored. We examined the survival and germinability of Bacillus subtilis spores exposed to simulated martian conditions that include solar radiation. Spores of B. subtilis that contain luciferase resulting from expression of an sspB-luxAB gene fusion were deposited on aluminum coupons to simulate deposition on spacecraft surfaces and exposed to simulated Mars atmosphere and solar radiation. The equivalent of 42 min of simulated Mars solar radiation exposure reduced spore viability by nearly 3 logs, while germination-induced bioluminescence, a measure of germination metabolism, was reduced by less than 1 log. The data indicate that spores can retain the potential to initiate germination-associated metabolic processes and produce biological signature molecules after being rendered nonviable by exposure to Mars solar radiation.  相似文献

Cometary Dust
This review presents our understanding of cometary dust at the end of 2017. For decades, insight about the dust ejected by nuclei of comets had stemmed from remote observations from Earth or Earth’s orbit, and from flybys, including the samples of dust returned to Earth for laboratory studies by the Stardust return capsule. The long-duration Rosetta mission has recently provided a huge and unique amount of data, obtained using numerous instruments, including innovative dust instruments, over a wide range of distances from the Sun and from the nucleus. The diverse approaches available to study dust in comets, together with the related theoretical and experimental studies, provide evidence of the composition and physical properties of dust particles, e.g., the presence of a large fraction of carbon in macromolecules, and of aggregates on a wide range of scales. The results have opened vivid discussions on the variety of dust-release processes and on the diversity of dust properties in comets, as well as on the formation of cometary dust, and on its presence in the near-Earth interplanetary medium. These discussions stress the significance of future explorations as a way to decipher the formation and evolution of our Solar System.  相似文献

The observed scaling relations imply that supermassive black holes (SMBH) and their host galaxies evolve together. Near-Eddington winds from the SMBH accretion discs explain many aspects of this connection. The wind Eddington factor \(\dot{m}\) should be in the range ~1–30. A factor \(\dot{m}\sim 1\) give black hole winds with velocities v~0.1c, observable in X-rays, just as seen in the most extreme ultrafast outflows (UFOs). Higher Eddington factors predict slower and less ionized winds, observable in the UV, as in BAL QSOs. In all cases the wind must shock against the host interstellar gas and it is plausible that these shocks should cool efficiently. There is detailed observational evidence for this in some UFOs. The wind sweeps up the interstellar gas into a thin shell and propels it outwards. For SMBH masses below a certain critical (Mσ) value, all these outflows eventually stall and fall back, as the Eddington thrust of the wind is too weak to drive the gas to large radii. But once the SMBH mass reaches the critical Mσ value the global character of the outflow changes completely. The wind shock is no longer efficiently cooled, and the resulting thermal expansion drives the interstellar gas far from the black hole, which is unlikely to grow significantly further. Simple estimates of the maximum stellar bulge mass M b allowed by self-limited star formation show that the SMBH mass is typically about 10?3 M b at this point, in line with observation. The expansion-driven outflow reaches speeds v out?1200 km?s?1 and drives rates \(\dot{M}_{\mathrm{out}}\sim 4000~\mathrm {M}_{\odot }\,\mathrm{yr}^{-1}\) in cool (molecular) gas, giving a typical outflow mechanical energy L mech?0.05L Edd, where L Edd is the Eddington luminosity of the central SMBH. This is again in line with observation. These massive outflows may be what makes galaxies become red and dead, and can have several other potentially observable effects. In particular they have the right properties to enrich the intergalactic gas with metals. Our current picture of SMBH-galaxy coevolution is still incomplete, as there is no predictive theory of how the hole accretes gas from its surroundings. Recent progress in understanding how large-scale discs of gas can partially cancel angular momentum and promote dynamical infall offers a possible way forward.  相似文献

Laminar dust flames in a reduced-gravity environment   总被引:1,自引:0,他引:1
The propagation of laminar dust flames in suspensions of iron in gaseous oxidizers was studied in a low-gravity environment onboard a parabolic flight aircraft. The reduction of buoyancy-induced convective flows and particle settling permitted the measurement of fundamental combustion parameters, such as the burning velocity and the flame quenching distance over a wide range of particle sizes and in different gaseous mixtures. Experimentally measured flame speeds and quenching distances were found in good agreement with theoretical predictions of a simplified analytical model that assumes particles burning in a diffusive mode. However, the comparison of flame speeds in oxygen–argon and oxygen–helium iron suspensions indicates the possibility that fine micron-sized particles burn in the kinetic mode. Furthermore, when the particle spacing is large compared to the scale of the reaction zone, a theoretical analysis suggests the existence of a new so-called discrete flame propagation regime. Discrete flames are strongly dependent on particle density fluctuations and demonstrate directed percolation behavior near flame propagation limits. The experimental observation of discrete flames in particle suspensions will require low levels of gravity over extended periods available only on orbital platforms.  相似文献

Structurally complex life and intelligence evolved late on Earth; models for the evolution of global temperature suggest that, due to the increasing solar luminosity, the future life span of the (eukaryote) biosphere will be "only" about another billion years, a short time compared to the approximately 4 Ga since life began. A simple stochastic model (Carter, 1983) suggests that this timing might be governed by the necessity to pass a small number, n, of very difficult evolutionary steps, with n < 10 and a best guess of n = 4, in order for intelligent observers like ourselves to evolve. Here I extend the model analysis to derive probability distributions for each step. Past steps should tend to be evenly spaced through Earth's history, and this is consistent with identification of the steps with some of the major transitions in the evolution of life on Earth. A complementary approach, identifying the critical steps with major reorganizations in Earth's biogeochemical cycles, suggests that the Archean-Proterozoic and Proterozoic-Phanerozoic transitions might be identified with critical steps. The success of the model lends support to a "Rare Earth" hypothesis (Ward and Brownlee, 2000): structurally complex life is separated from prokaryotes by several very unlikely steps and, hence, will be much less common than prokaryotes. Intelligence is one further unlikely step, so it is much less common still.  相似文献
