The effects of microgravity on Jurkat cells--a T-lymphoid cell line--was studied on a sounding rocket flight. An automated pre-programmed instrument permitted the injection of fluorescent labelled concanavalin A (Con A), culture medium and/or fixative at given times. An in-flight 1 g centrifuge allowed the comparison of the data obtained in microgravity with a 1 g control having the same history related to launch and re-entry. After flight, the cells fixed either at the onset of microgravity or after a or 12 minute incubation time with fluorescent concanavalin A were labelled for vimentin and actin and analysed by fluorescence microscopy. Binding of Con A to Jurkat cells is not influenced by microgravity, whereas patching of the Con A receptors is significantly lower. A significant higher number of cells show changes in the structure of vimentin in microgravity. Most evident is the appearance of large bundles, significantly increased in the microgravity samples. No changes are found in the structure of actin and in the colocalisation of actin on the inner side of the cell membrane with the Con A receptors after binding of the mitogen.  相似文献

We report initial measurements from the ULECA sensor of the Max-Planck-Institut/University of Maryland experiment on ISEE-1. ULECA is an electrostatic deflection — total energy sensor consisting of a collimator, deflection analyzer and an array of solid state detectors. The position of a given detector, which determines the energy per charge of an incident particle, together with the measured energy determine the particle's charge state. We find that a rich variety of phenomena are operative in the transthermal energy regime (10 keV/Q to 100 keV/Q) covered by ULECA. Specifically, we present observations of locally accelerated protons, alpha particles, and heavier ions in the magnetosheath and upstream of the Earth's bow shock. Preliminary analysis indicates that the behavior of these locally accelerated particles is most similar at the same energy per charge.  相似文献

The Solar Wind Electron Proton Alpha Monitor (SWEPAM) experiment provides the bulk solar wind observations for the Advanced Composition Explorer (ACE). These observations provide the context for elemental and isotopic composition measurements made on ACE as well as allowing the direct examination of numerous solar wind phenomena such as coronal mass ejections, interplanetary shocks, and solar wind fine structure, with advanced, 3-D plasma instrumentation. They also provide an ideal data set for both heliospheric and magnetospheric multi-spacecraft studies where they can be used in conjunction with other, simultaneous observations from spacecraft such as Ulysses. The SWEPAM observations are made simultaneously with independent electron and ion instruments. In order to save costs for the ACE project, we recycled the flight spares from the joint NASA/ESA Ulysses mission. Both instruments have undergone selective refurbishment as well as modernization and modifications required to meet the ACE mission and spacecraft accommodation requirements. Both incorporate electrostatic analyzers whose fan-shaped fields of view sweep out all pertinent look directions as the spacecraft spins. Enhancements in the SWEPAM instruments from their original forms as Ulysses spare instruments include (1) a factor of 16 increase in the accumulation interval (and hence sensitivity) for high energy, halo electrons; (2) halving of the effective ion-detecting CEM spacing from ∼5° on Ulysses to ∼2.5° for ACE; and (3) the inclusion of a 20° conical swath of enhanced sensitivity coverage in order to measure suprathermal ions outside of the solar wind beam. New control electronics and programming provide for 64-s resolution of the full electron and ion distribution functions and cull out a subset of these observations for continuous real-time telemetry for space weather purposes. Over four Venus years of low altitude nightside PVO magnetometer observations are used to establish a new upper limit for the magnetic moment of Venus. Improvements over previous studies include data coverage and new instrument calibration information. The upper limit on an internal dipole moment is determined to be 8.4 × 1010 T m3.  相似文献

The paper presents a physical mechanism of large-scale vortex electric field generation in the ionospheric E- and F-layers. It shows that the planetary-scale, synoptic short-period (from several second to several hours) and fast processes (with propagation velocity higher than 1 km/s) produce a planetary-scale internal vortex electric field. Its value may far exceed that of the dynamo-field generated in the same ionospheric layer by local wind motion. We found, that an ionospheric source of the vortex electric field is spatial inhomogeneity of the geomagnetic field.  相似文献

Within the European space platform program the EURECA is being established as a ground-based platform for short microgravity missions. The development towards a serviceable platform for longer, scientific missions is described. The plan of an advanced space-based platform for increasing payload demands is outlined. The platform design and the adaptation to scientific missions and servicing operations are investigated. The cost-effective utilization of the different platform types using new operational concepts is analyzed in parametric life cycle cost calculations for different payloads and mission scenarios.  相似文献

Star identification can be accomplished by several different available algorithms that identify the stars observed by a star tracker. However, efficiency and reliability remain key issues and the availability of new active pixel cameras requires new approaches. Two novel algorithms for recursive mode star identification are presented here. The first approach is derived by the spherical polygon search (SP-search) algorithm, it was used to access all the cataloged stars observed by the sensor field-of-view (FOV) and recursively add/remove candidate cataloged stars according to the predicted image motion induced by camera attitude dynamics. Star identification is then accomplished by a star pattern matching technique which identifies the observed stars in the reference catalog. The second method uses star neighborhood information and a catalog neighborhood pointer matrix to access the star catalog. In the recursive star identification process, and under the assumption of \"slow\" attitude dynamics, only the stars in the neighborhood of previously identified stars are considered for star identification in the succeeding frames. Numerical tests are performed to validate the absolute and relative efficiency of the proposed methods.  相似文献

Using empirical velocity distributions derived from UVCS and SUMER ultraviolet spectroscopy, we construct theoretical models of anisotropic ion temperatures in the polar solar corona. The primary energy deposition mechanism we investigate is the dissipation of high frequency (10-10000 Hz) ion-cyclotron resonant Alfvén waves which can heat and accelerate ions differently depending on their charge and mass. We find that it is possible to explain the observed high perpendicular temperatures and strong anisotropies with relatively small amplitudes for the resonant waves. There is suggestive evidence for steepening of the Alfvén wave spectrum between the coronal base and the largest heights observed spectroscopically. Because the ion-cyclotron wave dissipation is rapid, even for minor ions like O5+, the observed extended heating seems to demand a constantly replenished population of waves over several solar radii. This indicates that the waves are generated gradually throughout the wind rather than propagated up from the base of the corona. The possibility of an ocean within the icy shell of Jupiter's moon Europa has established that world as a primary candidate in the search for extraterrestrial life within our Solar System. This paper evaluates the potential to detect evidence for microbial life by comparing laboratory studies of terrestrial microorganisms with measurements from the Galileo Near Infrared Imaging Spectrometer (NIMS). If the interior of Europa at one time harbored life, some evidence may remain in the surface materials. Examination of laboratory spectra of terrestrial extremophiles measured at cryogenic temperatures reveals distorted, asymmetric nearinfrared absorption features due to water of hydration. The band centers, widths, and shapes of these features closely match those observed in the Europa spectra. These features are strongest in reddish-brown, disrupted terrains such as linea and chaos regions. Narrow spectral features due to amide bonds in the microbe proteins provide a means of constraining the abundances of such materials using the NIMS data. The NIMS data of disrupted terrains exhibit distorted, asymmetric near-infrared absorption features consistent with the presence of water ice, sulfuric acid octahydrate, hydrated salts, and possibly as much as 0.2 mg cm(-3) of carbonaceous material that could be of biological origin. However, inherent noise in the observations and limitations of spectral sampling must be taken into account when discussing these findings.  相似文献

Effect of the size of rhizospheric bacterial populations on germination of seeds and development of simple terrestrial "wheat plants--rhizospheric microorganisms--artificial soil" and "wheat plants-artificial soil" systems has been studied. Experiments demonstrated that within specify ranges in the inoculate, the rhizospheric bacteria are capable of increasing the yield of germinated seeds and stimulate the growth of plantlets. Germination of seeds inoculated with bacteria was either stimulated, or inhibited or remained at control levels depending on the amount of bacteria. Plant biomass growth and total photoassimilation has been found to depend on the amount of bacteria on the plant roots: the higher the amount of bacteria on plant roots, the smaller is the biomass of plants but the total photoassimilation is, higher. Thus, depending on the amount of bacteria on the roots of plants the system either increases the biomass of plants or increases the total photoassimilation, i.e. "pumps" carbon through itself involving bacteria.
