Exposure of fungal conidia (Aspergillus ochraceus) or spores of Bacillus subtilis to extreme dryness or vacuum induces DNA lesions, including strand breaks and the formation of DNA-protein cross-links. In wet cells only a small amount of protein is bound to DNA, but exposure to conditions of lowered water activity results in an increasing number of cross-links between DNA and proteins. In fungal conidia these cross-links are detected after selective iodination (125 J) of the DNA-bound proteins followed by gel electrophoresis and subsequent autoradiography. Another approach is the labelling of DNA with 32P by means of nick translation and the detection of differences in the electrophoretic mobility of DNA before and after digestion with proteinase K of proteins bound to DNA. 相似文献

In the context of prebiotic chemistry in space, some of the outer planetary objects display H, C, N and O rich chemistry similar to the one in the biosphere of Earth. Of particular interest are Saturn's moon, Titan; Neptune's moon, Triton; and Pluto where extreme cold conditions prevail. Identifications of chemical species on these objects (surfaces and atmospheres) is essential to a better understanding of the radiation induced chemical reactions occurring thereon. There have been several ground based observations of these planetary objects in the infrared windows from 1 to 2.5 micrometers. Voyager also provided spectra in the thermal infrared (6 to 50 micrometers) region. Interpretation of these data require laboratory infrared spectra of relevant species under the temperature conditions appropriate to these objects. The results of some of these studies carried out in our laboratory and elsewhere and their impact on the analyses of the observed data will be summarized. 相似文献

We investigated the effect of substratum adhesiveness on stimulated lymphocyte blastogenesis by reducing and blocking cell adhesion with poly (2-hydroxyethyl methacrylate) (poly-HEMA) in a simple on-ground system. Cells grown on medium-thick and thick poly-HEMA films were rounded in shape and displayed no signs of spreading. By contrast, on tissue culture plastic and very thin poly-HEMA films, they showed clear signs of spreading. The mitogenic response of lymphocytes grown on thick poly-HEMA films was reduced by up to 68% of the control (tissue culture plastic). Interferon-gamma production was near zero when the cells were grown on the least adhesive substratum. On uncoated plastic, activated lymphocytes subjected to high gravity (20g) exhibited an increased proliferation rate (40%) compared with 1g. By contrast, on poly-HEMA, high gravity did not improve lymphocyte responsiveness. These results show that activated lymphocytes need to anchor and spread prior to achieving an optimal proliferation response. We conclude that decreased lymphocyte adhesion could contribute to the depressed in vitro lymphocyte responsiveness found in the microgravity conditions of space flight. 相似文献

The possibility of constructing an optical scheme of a telescope for a small satellite based on mirrors with freeform surfaces and a curved photodetector is considered. It is shown that the use of a new element base allows the effective area of the main mirror to be maximized and the field of view up to 9.4 times to be increased in comparison with the classical scheme of the Ritchey–Chretien scheme in ensuring the high image quality. 相似文献

The water content of magma oceans is widely accepted as a key factor that determines whether a terrestrial planet is habitable. Water ocean mass is determined as a result not only of water delivery and loss, but also of water partitioning among several reservoirs. Here we review our current understanding of water partitioning among the atmosphere, magma ocean, and solid mantle of accreting planetary embryos and protoplanets just after giant collisions. Magma oceans are readily formed in planetary embryos and protoplanets in their accretion phase. Significant amounts of water are partitioned into magma oceans, provided the planetary building blocks are water-rich enough. Particularly important but still quite uncertain issues are how much water the planetary building blocks contain initially and how water goes out of the solidifying mantle and is finally degassed to the atmosphere. Constraints from both solar-system explorations and exoplanet observations and also from laboratory experiments are needed to resolve these issues. 相似文献
