The search for unequivocal signs of life on other planetary bodies is one of the major challenges for astrobiology. The failure to detect organic molecules on the surface of Mars by measuring volatile compounds after sample heating, together with the new knowledge of martian soil chemistry, has prompted the astrobiological community to develop new methods and technologies. Based on protein microarray technology, we have designed and built a series of instruments called SOLID (for "Signs Of LIfe Detector") for automatic in situ detection and identification of substances or analytes from liquid and solid samples (soil, sediments, or powder). Here, we present the SOLID3 instrument, which is able to perform both sandwich and competitive immunoassays and consists of two separate functional units: a Sample Preparation Unit (SPU) for 10 different extractions by ultrasonication and a Sample Analysis Unit (SAU) for fluorescent immunoassays. The SAU consists of five different flow cells, with an antibody microarray in each one (2000 spots). It is also equipped with an exclusive optical package and a charge-coupled device (CCD) for fluorescent detection. We demonstrated the performance of SOLID3 in the detection of a broad range of molecular-sized compounds, which range from peptides and proteins to whole cells and spores, with sensitivities at 1-2?ppb (ng?mL?1) for biomolecules and 10? to 103 spores per milliliter. We report its application in the detection of acidophilic microorganisms in the Río Tinto Mars analogue and report the absence of substantial negative effects on the immunoassay in the presence of 50?mM perchlorate (20 times higher than that found at the Phoenix landing site). Our SOLID instrument concept is an excellent option with which to detect biomolecules because it avoids the high-temperature treatments that may destroy organic matter in the presence of martian oxidants.  相似文献

Prominence seismology is a rapidly developing topic which seeks to infer the internal structure and properties of solar prominences from the study of its oscillations. Two-dimensional high-resolution observations suggest that filaments can be considered as made by small scale fibrils, having a cool region, stacked one after another in the vertical and horizontal directions. An extense observational background about oscillations in filaments has been gathered during the last 20 years and these observations point out that fibrils or groups of fibrils can oscillate independently. From the theoretical point of view, small amplitude oscillations in single and multifibril configurations have been studied as a first step to explain observational features.  相似文献

We describe recent progress in physics-based models of the plasmasphere using the fluid and the kinetic approaches. Global modeling of the dynamics and influence of the plasmasphere is presented. Results from global plasmasphere simulations are used to understand and quantify (i) the electric potential pattern and evolution during geomagnetic storms, and (ii) the influence of the plasmasphere on the excitation of electromagnetic ion cyclotron (EMIC) waves and precipitation of energetic ions in the inner magnetosphere. The interactions of the plasmasphere with the ionosphere and the other regions of the magnetosphere are pointed out. We show the results of simulations for the formation of the plasmapause and discuss the influence of plasmaspheric wind and of ultra low frequency (ULF) waves for transport of plasmaspheric material. Theoretical models used to describe the electric field and plasma distribution in the plasmasphere are presented. Model predictions are compared to recent Cluster and Image observations, but also to results of earlier models and satellite observations.  相似文献

In this review we present the main results obtained by the ISO satellite on the abundance and spatial distribution of water vapor in the direction of molecular clouds, evolved stars, galaxies, and in the bodies of our Solar System. We also discuss the modeling of H2O and the difficulties found in the interpretation of the data, the need of collisional rates and the perspectives that future high angular and high spectral resolution observations of H2O with the Herschel Space Observatory will open.  相似文献

The next time humans set foot on the Moon or another planet, will we treat the crew like we would a sample return mission when they come back to Earth? This may seem a surprising or even provocative question, but it is one we need to address. The hurdles and hazards of sending humans to Mars – for example, the technology constraints and physiological and psychological challenges – are many; but let us not forget the need to protect populations and environments from the risk of contamination [United Nations, treaty on principles governing the activities of states in the exploration and use of outer space, including the Moon and other celestial bodies (the “Outer Space Treaty”) referenced 610 UNTS 205 - resolution 2222(XXI) of December 1966].  相似文献

What hazards might biological contamination pose to planets, comets and other celestial bodies visited by probes launched from Earth? What hazards might returning probes pose to Earth and its inhabitants? What should be considered an acceptable level of risk? What technologies, procedures and constraints should be applied? What sort of attitude has to be chosen concerning human crews, who themselves could become both contaminated victims and contaminating agents? The vast issue of planetary protection must, more than ever, spark ethical debate. Space treaty, COSPAR recommendations offer borders and context for this reflection, which has to be introduced in the actual humanist: never has been anthropocentrism so practical and concerned, in the same time, by the next generations, because of the historical character of life. At least an ethics of risk is necessary (far from the myth of zero-risk) for all the three types of contamination: other celestial bodies (forward contamination), Earth (backward contamination) and astronauts.  相似文献
