Biogenic stromatolites are sources of significant information on the evolution of microbial life. Despite their evolutionary significance, little is known about the mechanisms of osmoadaptation by microorganisms that comprise living stromatolites thriving in hypersaline environments. Osmoadaptive strategies for Halococcus hamelinensis, a novel halophilic archaeon recently isolated from living stromatolites in the hypersaline reaches of Shark Bay, were thus a particular interest in this study. To investigate the possibility of "salt-in-cytoplasm"-associated osmoadaptation for this archaeon, flame photometry studies were performed. From the results, it was evident that this halophilic archaeon did not accumulate intracellular K(+) ions when cells were exposed to either osmotic shock or conditions with gradual increments in salinity. These results were further supported by polymerase chain reaction (PCR) analyses where there was no evidence for the existence of homologous genes to an ATP-driven, high-affinity potassium uptake system in Halococcus hamelinensis. To identify an alternative salt adaptation mechanism associated with accumulation of compatible solutes for this archaeon, (1)H nuclear magnetic resonance (NMR) spectroscopy experiments were carried out. Results indicate that glycine betaine, trehalose, and glutamate are solutes likely to be involved in osmoregulation in this archeaon. Subsequent (1)H NMR analysis of cell extracts from this microorganism grown under various NaCl concentrations revealed that intracellular levels of glycine betaine increased with increasing concentrations of NaCl. This behavior of increasing glycine betaine concentration with increasing external NaCl is consistent with its identity as an osmolyte. In contrast, intracellular levels of trehalose were decreased in high concentrations of NaCl. This provides evidence that compatible solute accumulation appears to be the preferential salt regulation mechanism for this haloarchaeon, in contrast to the salt-in-cytoplasm strategy employed by many other halophilic archaea.  相似文献

Pitch Lake in Trinidad and Tobago is a natural asphalt reservoir nourished by pitch seepage, a form of petroleum that consists of mostly asphaltines, from the surrounding oil-rich region. During upward seepage, pitch mixes with mud and gases under high pressure, and the lighter portion evaporates or is volatilized, which produces a liquid asphalt residue characterized by low water activity, recalcitrant carbon substrates, and noxious chemical compounds. An active microbial community of archaea and bacteria, many of them novel strains (particularly from the new Tar ARC groups), totaling a biomass of up to 10(7) cells per gram, was found to inhabit the liquid hydrocarbon matrix of Pitch Lake. Geochemical and molecular taxonomic approaches revealed diverse, novel, and deeply branching microbial lineages with the potential to mediate anaerobic hydrocarbon degradation processes in different parts of the asphalt column. In addition, we found markers for archaeal methane metabolism and specific gene sequences affiliated with facultative and obligate anaerobic sulfur- and nitrite-oxidizing bacteria. The microbial diversity at Pitch Lake was found to be unique when compared to microbial communities analyzed at other hydrocarbon-rich environments, which included Rancho Le Brea, a natural asphalt environment in California, USA, and an oil well and a mud volcano in Trinidad and Tobago, among other sites. These results open a window into the microbial ecology and biogeochemistry of recalcitrant hydrocarbon matrices and establish the site as a terrestrial analogue for modeling the biotic potential of hydrocarbon lakes such as those found on Saturn's largest moon Titan.  相似文献

A new adaptive nonlinear guidance law is proposed here. The fourth order state equation for integrated guidance and control loop is formulated taking into consideration the target uncertainties and control loop dynamics. The state equation is further changed into the normal form by nonlinear coordinate transformation. Using the normal form of state equation, an adaptive nonlinear guidance law is proposed to compensate for the uncertainties in both target acceleration and control loop dynamics. The proposed law adopts the sliding mode control approach with adaptation for unknown bound of uncertainties. The present approach can effectively solve the existing guidance problem against target maneuver and the limited performance of control loop. We have provided the stability analyses and performed simulations comparing favorably our approach to the state of the art.  相似文献

This paper deals with data association using three sets of passive linear array sonars (PLAS) geometrically positioned in a Y-shaped configuration, fixed in an underwater environment. The data association problem is directly transformed into a 3D assignment, which is known to be NP hard. For generic passive sensors, it can be solved using conventional algorithms, while in PLAS, it becomes a formidable task due to the presence of bearing ambiguity. Thus, the central issue of the problem in PLAS is how to eliminate the bearing ambiguity without increasing tracking error. To solve this problem, the 3D assignment algorithm used the likelihood value of only those observed bearing measurements is modified by incorporating frequency information in consecutive time-aligned scans. The region of possible ghost targets is first established by the geometrical relation of PLAS with respect to target. The ghost targets are then confirmed and eliminated by generating multiple observations in consecutive scans. Representative simulations demonstrate the effectiveness of the proposed approach.  相似文献

This paper documents the design of a laser/radio frequency (RF) Soldier Identification (ID) System developed by Dynetics, Inc., Harris, Corp., and the US Army Communications and Electronics Command (CECOM). The Soldier ID system includes an Interrogation Unit with a programmable activation code. The Interrogation Unit consists of a directive, eye-safe laser and a spread-spectrum RF transceiver. This allows for a low probability of intercept (LPI) interrogation, which is of interest during covert operations. A Responder Unit is worn, for example, by a soldier and transmits an LPI spread-spectrum RF response, only after receiving the proper interrogation codes. The basic subsystems for the identification system are a Laser Interrogation Unit, an RF Responder Unit, and an electronic Programming Unit. The operating principles for the subsystem are reviewed, and the design issues are discussed. In addition to the preliminary design performed under Phase I of the program, a breadboard system was developed to validate the proof-of-principle concept. Hardware implementation is reviewed and test results are presented. The brassboard development and engineering plans are also presented  相似文献

Sustaining weapons system hardware and software represents a significant and ever-increasing portion of total system cost. Hardware components are becoming obsolete much sooner while weapons system lifetimes are increasing, We must identify more cost-effective solutions to engineering and reengineering these subsystems. Verifying and validating weapons systems are two of the most costly parts of either engineering process. Traditionally, hardware validation and verification is done by simulation and testing, In the past few years, math- and logic-based formal methods tools have begun to scale up to and be applied successfully to real-world problems. Incorporating formal verification methods into engineering and reengineering processes will cost-effectively and significantly improve the level of trust and the quality of our weapons systems. Formal methods are especially well suited for redesigning current weapon systems which have become unsupportable due to component obsolescence because they help minimize the astronomical costs of rigorously reverifying the reengineered components. We believe that formal methods are an important tool for effective engineering of future weapon systems  相似文献

A robust adaptive control scheme is proposed that can be applied to a practical autopilot design for feedback-linearized skid-to-turn (STT) missiles with aerodynamic uncertainties. The approach is to add a robust adaptive controller to a feedback-linearizing controller in order to reduce the influence of the aerodynamic uncertainties. The proposed robust adaptive control scheme is based on a sliding mode control technique with an adaptive law for estimating the unknown upper bounds of uncertain parameters. A feature of the proposed scheme is that missile systems with aerodynamic uncertainties can be controlled effectively over a wide operating range of flight conditions. It is shown, using Lyapunov stability theory, that the proposed scheme can give sufficient tracking capability and stability for a feedback-linearized STT missile with aerodynamic uncertainties. The six-degree-of-freedom nonlinear simulation results also show that good performance for several uncertainty models and engagement scenarios can be achieved by the proposed scheme in practical night conditions  相似文献

A new form of the probabilistically strongest neighbor filter (PSNF) algorithm taking into account the number of validated measurements is proposed. The probabilistic nature of the strongest neighbor (SN) measurement in a cluttered environment is shown to be varied with respect to the number of validated measurements. Incorporating the number of validated measurements into design of the PSNF produces a consistent and cost effective data association method. Simulation studies show that the new filter is less sensitive to the unknown spatial clutter density and is more reliable for practical target tracking in nonhomogeneous clutter than the existing PSNF. It has similar performances to the probabilistic data association filter amplitude information (PDAF-AI) with much less computational complexities.  相似文献

This paper presents a new algorithm for three-axis attitude determination using measurement vectors composed of incomplete vector components. Using two separate algorithms, called Kalman's recursive and Vaughan's nonrecursive, exact closed-form solutions of the steady-state two-state exponentially correlated velocity (ECV) and three-state exponentially correlated acceleration (ECA) target-tracking filters are derived for discrete position measurements. For this particular application, the superiority of Vaughan's nonrecursive over Kalman's recursive algorithm is observed.  相似文献
