Because of their rapid development, amphibians have been important model organisms in studies of how microgravity affects vertebrate growth and differentiation. Both urodele (salamanders) and anuran (frogs and toads) embryos have been raised in orbital flight, the latter several times. The most commonly reported and striking effects of microgravity on tadpoles are not in the vestibular system, as one might suppose, but in their lungs and tails. Pathological changes in these organs disrupt behavior and retard larval growth. What causes malformed (typically lordotic) tadpoles in microgravity is not known, nor have axial pathologies been reported in every flight experiment. Lung pathology, however, has been consistently observed and is understood to result from the failure of the animals to inflate their lungs in a timely and adequate fashion. We suggest that malformities in the axial skeleton of tadpoles raised in microgravity are secondary to problems in respiratory function. We have used high speed videography to investigate how tadpoles breathe air in the 1G environment. The video images reveal alternative species-specific mechanisms, that allow tadpoles to separate air from water in less that 150 ms. We observed nothing in the biomechanics of air-breathing in 1G that would preclude these same mechanisms from working in microgravity. Thus our kinematic results suggest that the failure of tadpoles to inflate their lungs properly in microgravity is due to the tadpoles' inability to locate the air-water interface and not a problem with the inhalation mechanism per se. 相似文献

We report measurements of chromosomal aberrations in peripheral blood lymphocytes from cancer patients undergoingradiotherapy treatment. Patients with cervix or esophageal cancer were treated with 10 MV X-rays produced at a LINAC accelerator, or high-energy carbon ions produced at the HIMAC accelerator at the National Institute for Radiological Sciences (NIRS) in Chiba. Blood samples were obtained before, during, and after the radiation treatment. Chromosomes were prematurely condensed by incubation in calyculin A. Aberrations in chromosomes 2 and 4 were scored after fluorescence in situ hybridization with whole-chromosome probes. Pre-treatment samples were exposed in vitro to X-rays, individual dose-response curves for the induction of chromosomal aberrations were determined, and used as calibration curves to calculate the effective whole-body dose absorbed during the treatment. This calculated dose, based on the calibration curve relative to the induction of reciprocal exchanges, has a sharp increase after the first few fractions of the treatment, then saturates at high doses. Although carbon ions are 2–3 times more effective than X-rays in tumor sterilization, the effective dose was similar to that of X-ray treatment. However, the frequency of complex-type chromosomal exchanges was much higher for patients treated with carbon ions than X-ray. 相似文献

Space flight results in the exposure of astronauts to a mixed field of radiation composed of energetic particles of varying energies, and biological indicators of space radiation exposure provides a better understanding of the associated long-term health risks. Current methods of biodosimetry have employed the use of cytogenetic analysis for biodosimetry, and more recently the advent of technological progression has led to advanced research in the use of genomic and proteomic expression profiling to simultaneously assess biomarkers of radiation exposure. We describe here the technical advantages of the Luminex(TM) 100 system relative to traditional methods and its potential as a tool to simultaneously profile multiple proteins induced by ionizing radiation. The development of such a bioassay would provide more relevant post-translational dynamics of stress response and will impart important implications in the advancement of space and other radiation contact monitoring. 相似文献

Carcinogenesis is postulated to be a progressive multistage process characterized by an increase in genomic instability and clonal selection with each mutational event endowing a selective growth advantage. Genomic instability as manifested by the amplification of specific gene fragments is common among tumor and transformed cells. In the present study, immortalized human bronchial (BEP2D) cells were irradiated with graded doses of either 1GeV/nucleon 56Fe ions or 150 keV/μm alpha particles. Transformed cells developed through a series of successive steps before becoming tumorigenic in nude mice. Tumorigenic cells showed neither ras mutations nor deletion in the p16 tumor suppressor gene. In contrast, they harbored mutations in the p53 gene and over-expressed cyclin D1. Genomic instability among transformed cells at various stage of the carcinogenic process was examined based on frequencies of PALA resistance. Incidence of genomic instability was highest among established tumor cell lines relative to transformed, non-tumorigenic and control cell lines. Treatment of BEP2D cells with a 4 mM dose of the aminothiol WR-1065 significantly reduced their neoplastic transforming response to 56Fe particles. This model provides an opportunity to study the cellular and molecular mechanisms involved in malignant transformation of human epithelial cells by heavy ions. 相似文献

The region of South Atlantic Geomagnetic Anomaly (SAGA) was investigated by the Intercosmos-Bulgaria-1300 satellite, launched on August 7, 1981. On the basis of data obtained from 15 orbits during increased geomagnetic activity in August 1981, a map of the Anomaly was elaborated. Two centres of activity were identified. Early Earth and early Mars were similar enough such that past geochemical and climatic conditions on Mars may have also been favorable for the origin of life. However, one of the most striking differences between the two planets was the low partial pressure of dinitrogen (pN2) on early Mars (18 mb). On Earth, nitrogen is a key biological element and in many ecosystems the low availability of fixed nitrogen compounds is the main factor limiting growth. Biological fixation of dinitrogen on Earth is a crucial source of fixed nitrogen. Could the low availability of dinitrogen in the primordial Martian atmosphere have prevented the existence, or evolution of Martian microbiota? Azotobacter vinelandii and Azomonas agilis were grown in nitrogen free synthetic medium under various partial pressures of dinitrogen ranging from 780-0 mb (total atmosphere=1 bar). Below 400 mb the biomass, cell number, and growth rate decreased with decreasing pN2. Both microorganisms were capable of growth at a pN2 as low as 5 mb, but no growth was observed at a pN2 < or = 1 mb. The data appear to indicate that biological nitrogen fixation could have occurred on primordial Mars (pN2=18 mb) making it possible for a biotic system to have played a role in the Martian nitrogen cycle. It is possible that nitrogen may have played a key role in the early evolution of life on Mars, and that later a lack of available nitrogen on that planet (currently, pN2=0.2 mb) may have been involved in its subsequent extinction. 相似文献

The complex problems associated with the aerodynamics of blade/vortex interactions, the dynamics of a blade displaced out of its plane of rotation, and the position control of a hovering helicopter are discussed. Simplified models help to describe, and appear to be adequate for analysing, many aspects of these problems. Free wake analyses are shown to be of importance in determining blade loads and performance for hovering flight or for wind turbines. Substorm evolution of the near-Earth (|X|<15 RE) plasma sheet has been emphasized recently because the inner tail is thought to link closely to the substorm auroral activity in the ionosphere during the early stage of substorms. In this paper, we discuss how the inner tail substorm phenomena during the late substorm growth phase and early expansion phase are accounted for by the two prevailing substorm models, namely, the near-Earth neutral line model and the current disruption model. We find that the late growth phase features are more satisfactorily accounted for by the current disruption model than by the near-Earth neutral line model. In addition, detailed observations on current disruption show evidence inconsistent with the proposed idea of dipolarization being due to plasma flow braking from reconnection in the mid-tail region, which poses a difficulty to the near-Earth neutral line model as well. 相似文献

It has generally been assumed that a geomagnetic storm is entirely driven by external forces—e.g., solar wind Ey = Vx × Bz, Vx, V2x (where the components of the electric field, E, the magnetic field, B, and velocity, V, are given in GSE coordinates)—which would imply that particle injections in the ring current (RC) or outer radiation belts should be highly correlated. However the data from ISTP are showing that the magnetosphere can have at least two very different responses to the same solar wind (SW) conditions: a classic, enhanced RC with Dst response, or a 1000-fold increase in the outer radiation belt MeV electrons (ORBE). August 29, October 14 and 23, 1996 are examples of Dst storms, whereas April 15, 1996 and January 10, 1997 are examples of MeV storms. It is this second response that is so deadly to some geosynchronous spacecraft, whereas geomagnetic storms are categorized by the first response. Neither of these appear to be correlated to the SW conditions driving substorms. Why should the SW energy appear in the radiation belts or the ring current independently? We hypothesize that the RC couples to the electric power available (Ey), the ORBE couple to the mechanical power available (Vx), and the Tail couples to the magnetic energy (Bz) available in the SW. The transducer for RC may be subauroral parallel potentials, the transducer for ORBE may be the cusp, while the Tail substorm transducer is yet a third independent mechanism for extracting SW energy. Evidence for this theory comes from the novel POLAR satellite that traverses the cusp, the plasmasheet and the radiation belts. 相似文献
