Theories of cellular radiation sensitivity that preclude a significant role for cellular repair processes in the final biological expression of cellular damage induced by ionizing radiation are unsound. Experiments are discussed here in which the cell-cycle dependency of the repair deficiency of the S/S variant, of the L5178Y murine leukemic lymphoblast was examined by treatment with the heavy ions, 20Ne, 28Si, 40Ar, 56Fe and 93Nb. Evidence from those studies, which will be described in detail elsewhere, provide support for the notion that as the linear energy transfer (LET infinity) of the incident radiation increases the ability of the S/S cell to repair radiation damage decreases until effectively it is eliminated around 500 keV/micrometer. In the region of the latter LET infinity value, the behavior of the S/S cell approximates the ideal case of target theory where post-irradiation metabolism (repair) does not influence cell survival. The expression of this phenomenon among different cell types and tissues will depend upon the actual repair systems involved and other considerations.  相似文献

The effect of elevated temperatures of 35 and 45 degrees C (at the intensities of photosynthetically active radiation 322, 690 and 1104 micromoles m-2 s-1) on the photosynthesis, respiration, and qualitative and quantitative composition of the volatiles emitted by wheat (Triticum aestuvi L., cultivar 232) crops was investigated in growth chambers. Identification and quantification of more than 20 volatile compounds (terpenoids--alpha-pinene, delta 3 carene, limonene, benzene, alpha- and trans-caryophyllene, alpha- and gamma-terpinene, their derivatives, aromatic hydrocarbons, etc.) were conducted by gas chromatograph/mass spectrometry. Under light intensity of 1104 micromoles m-2 s-1 heat resistance of photosynthesis and respiration increased at 35 degrees C and decreased at 45 degrees C. The action of elevated temperatures brought about variations in the rate and direction of the synthesis of volatile metabolites. The emission of volatile compounds was the greatest under a reduced irradiation of 322 micromoles m-2 s-1 and the smallest under 1104 micromoles m-2 s-1 at 35 degrees C. During the repair period, the contents and proportions of volatile compounds were different from their initial values, too. The degree of disruption and the following recovery of the functional state depended on the light intensity during the exposure to elevated temperatures. The investigation of the atmosphere of the growth chamber without plants has revealed the substances that were definitely technogenic in origin: tetramethylurea, dimethylsulfide, dibutylsulfide, dibutylphthalate, and a number of components of furan and silane nature.  相似文献

Transformation of the mechanical input in the chain: acceleration of otolithic membrane (OM)-displacement of the OM gel layer -deflection of hair cell bundle (HCB) -deformation of the system of tip-links- formation of temporal pattern of polarization was studied using simplified analytical models of these stages of conversion of mechanical stimulus into the HCB electrical response. The process of transformation of information in this chain was considered for two extreme cases of OM gel-HCB interaction: 1) the HCBs exactly follow the gel displacement; 2) stiff stereocilia and weak surrounding gel allow the relative motion of the bundle with regard to the gel. The analysis of a simplified model of cell polarization based on threshold triggering of the HCB tip-links allows to hypothesize that spatially nonhomogeneous HCB structure with a set of stereocilia of varying heights is designed to perceive spatially nonhomogeneous gel displacements caused by external acceleration. Thus, the HCB-OM gel interaction in the first case leads to formation of temporal pattern of depolarization that corresponds to the temporal pattern of gel displacement. In the second case the kinetics of depolarization reflects time dependence of gel displacement velocity.  相似文献

We have modeled “gradual” solar energetic particle events through numerical simulations using a StochasticDifferential Equation (SDE) method. We consider that energetic particle events are roughly divided into two groups: (1) where the shock was driven by coronal mass ejections (CMEs) associated with large solar flares, and (2) where they have no related solar events apart from the CMEs. (The detailed classification of energetic particle events was discussed in our previous paper.) What we call “gradual” solar energetic particle events belong to the former group. Particles with energies greater than 10 MeV are observed within several hours after the occurrence of flares and CMEs in many gradual events. By applying the SDE method coupled with particle splitting to diffusive acceleration, we found that an injection of high energy particles is necessary for early enhancement of such a high-energy proton flux and that it should not be presumed that the solar wind particles act as the seed population.  相似文献

The effects of microgravity on Jurkat cells--a T-lymphoid cell line--was studied on a sounding rocket flight. An automated pre-programmed instrument permitted the injection of fluorescent labelled concanavalin A (Con A), culture medium and/or fixative at given times. An in-flight 1 g centrifuge allowed the comparison of the data obtained in microgravity with a 1 g control having the same history related to launch and re-entry. After flight, the cells fixed either at the onset of microgravity or after a or 12 minute incubation time with fluorescent concanavalin A were labelled for vimentin and actin and analysed by fluorescence microscopy. Binding of Con A to Jurkat cells is not influenced by microgravity, whereas patching of the Con A receptors is significantly lower. A significant higher number of cells show changes in the structure of vimentin in microgravity. Most evident is the appearance of large bundles, significantly increased in the microgravity samples. No changes are found in the structure of actin and in the colocalisation of actin on the inner side of the cell membrane with the Con A receptors after binding of the mitogen.  相似文献

In this paper we discuss the effect of microgravity on T cells and we present the data of studies with two new machines for 0 g simulations. Several experiments in space show that mitogenic T cell activation is lost at 0 g. Immunocytochemistry indicates that such effect is associated with changes of the cytoskeleton. Biochemical studies suggest that the lack of expression of the interleukin-2 receptor is one of the major causes of the loss of activity. In fact, interleukin-2 is the third signal required for full activation. In order to deepen our investigations we are now working with the free-fall machine, FFM, invented by D. Mesland, and with the random positioning machine, RPM, or three-dimensional clinostat, developed by T. Hoson. The FFM produces periods of free-fall lasting approximately 800 ms followed by bounces of 15-30 g lasting 45-60 ms. The RPM eliminates the effect of gravity by rotating biological specimen randomly around two orthogonal axes. While the FFM failed to reproduce the results obtained with T lymphocytes in space, the data from the RPM are in good agreement with those in real microgravity. In fact, the inhibition of the mitotic index in the RPM is 89% compared to static controls. The RPM (as the FFM) can carry markedly larger specimen than the fast rotating clinostat and thus allows to conduct comprehensive studies to select suitable biological objects for further investigations in space.  相似文献

The equilibrium between the rods and panels in the ribbed shells is considered based on the total system of structural mechanics equations as applied to a discrete model. Also shown is an approach to solving the problems that permits the stiffness matrix of shear panels to be obtained with regard for the equilibrium along the boundary with rods. The connection with the standard hybrid method of constructing stiffness matrices is indicated. The solutions for separate elements and examples illustrating the approach being proposed are presented.  相似文献

In this paper, we consider a problem of synthesizing the control system of a small gliding (unpowered) aerial vehicle (GAV) with a high-aspect ratio wing (?? ?? 10).
