The effects of hypergravity on growth and osmoregulation were examined in dark-grown azuki bean epicotyls. Elongation growth of epicotyls was promptly suppressed by hypergravity at 300g. On the contrary, the increase in fresh weight of epicotyls during incubation was not suppressed by hypergravity at 300g at least up to 6 h. Also, the level of total osmotic solutes increased during epicotyl growth for 6 h, which was not affected by hypergravity. These results suggest that azuki bean epicotyls are capable of maintaining osmoregulation even under 300g conditions for a short period. On the other hand, the increase in fresh weight of epicotyls was suppressed, in addition to suppression of elongation growth, when seedlings were treated with 300g for 24 h. The increase in level of total osmotic solutes was also inhibited by 24 h hypergravity treatment, which was accounted by the reduced levels of organic solutes, such as sugars and amino acids. Furthermore, the dry weight of seeds decreased during incubation for 24 h, but the decrease was inhibited by hypergravity at 300g. Hypergravity treatment at 300g for 24 h also increased the pH value of apoplastic solution in epicotyls. Taken together, these results suggest that the translocation of organic solutes from the seed to epicotyls is inhibited by prolonged hypergravity treatment, which may underlie the suppression of epicotyl growth, and that the breakdown of H+ gradient across the plasma membrane in epicotyl cells may be at least partly involved in the reduction of organic solute accumulation under hypergravity conditions.  相似文献

Seedlings of azuki bean (Vigna angularis Ohwi et Ohashi) were cultivated under hypergravity conditions, and changes in membrane lipid composition in their epicotyls were analyzed. Under hypergravity conditions at 300g, the levels of total sterols, phospholipids, and fatty acids per fresh weight were kept higher, as compared with 1g controls. In particular, sterol levels were prominently increased by hypergravity. On the other hand, hypergravity did not clearly influence the levels of each phospholipid and glycolipid class, or their fatty acid compositions. Thus, the effect of hypergravity on membrane lipid metabolism was specific for sterol biosynthesis. In various regions of azuki epicotyls, high growth rate was associated with high sterol levels. Hypergravity suppressed elongation growth and stimulated lateral expansion of azuki epicotyls. In the presence of lovastatin, an inhibitor of sterol biosynthesis, at 30 μM, such changes in growth parameters occurred even under 1g conditions, suggesting that lovastatin made epicotyls hypersensitive to the gravitational force. These results support the hypothesis that membrane sterols are involved in maintenance of normal growth capacity of plant organs against gravity.  相似文献

Simulated microgravity (SMG) can inhibit proliferation and enhance microcystin production of Microcystis aeruginosa. We investigated the role of nitric oxide (NO) in regulating the SMG induced changes of proliferation, photochemical system II photochemical activity, pigment, soluble protein and microcystin production in M. aeruginosa. M. aeruginosa was exposed to 0.1 mM sodium nitroprusside (SNP, NO donor) or 0.02 mM 2-(4-carboxyphenyl)-4, 4, 5, 5-tetramethylimidazoline-1-oxyl-3-oxide (c-PTIO, NO scavenger) alone or in combination with SMG for 48 h. SMG and SNP inhibited the growth of M. aeruginosa while c-PTIO had no effect on cell number. As to yield, the negative effect of SMG was augmented by SNP and suppressed by c-PTIO. The intracellular concentrations of chlorophyll a, carotenoid, phycocyanin, soluble protein and microcystin were increased by SMG after 48 h. The effects of SMG on these metabolic processes could be enhanced by SNP and be partly eliminated by c-PTIO. Moreover, SNP and c-PTIO only functioned in these biochemical processes under SMG, unlike in the regulation of cell proliferation and yield. These results showed that the effects of SMG could be enhanced by adding exogenous NO and be mitigated by scavenging endogenous NO, revealing the involvement of NO in the changes in biochemistry processes induced by SMG in M. aeruginosa.  相似文献

The effects of gravity on bone metabolism are unclear, and little has been reported about the effects of hypergravity on the mature skeleton. Since low gravity has been shown to decrease bone volume, we hypothesized that hypergravity increases bone volume. To clarify this hypothesis, adult female rats were ovariectomized and exposed to hypergravity (2.9G) using a centrifugation system. The rats were killed 28 days after the start of loading, and the distal femoral metaphysis of the rats was studied. Bone architecture was assessed by micro-computed tomography (micro-CT) and bone mineral density was measured using peripheral quantitative CT (pQCT). Hypergravity increased the trabecular bone volume of ovariectomized rats. Histomorphometric analyses revealed that hypergravity suppressed both bone formation and resorption and increased bone volume in ovariectomized rats.  相似文献

Plants grown on long-term space missions will likely be grown in low pressure environments (i.e., hypobaria). However, in hypobaria the transpiration rates of plants can increase and may result in wilting if the water is not readily replaced. It is possible to reduce transpiration by increasing the partial pressure of CO2 (pCO2), but the effects of pCO2 at high levels (>120 Pa) on the growth and transpiration of plants in hypobaria are not known. Therefore, the effects of pCO2 on the growth and transpiration of radish (Raphanus sativus var. Cherry Bomb II) in hypobaria were studied. The fresh weight (FW), leaf area, dry weight (DW), CO2 assimilation rates (CA), dark respiration rates (DR), and transpiration rates from 26 day-old radish plants that were grown for an additional seven days at different total pressures (33, 66 or 101 kPa) and pCO2 (40 Pa, 100 Pa and 180 Pa) were measured. In general, the dry weight of plants increased with CO2 enrichment and with lower total pressure. In limiting pCO2 (40 Pa) conditions, the transpiration for plants grown at 33 kPa was approximately twice that of controls (101 kPa total pressure with 40 Pa pCO2). Increasing the pCO2 from 40 Pa to 180 Pa reduced the transpiration rates for plants grown in hypobaria and in standard atmospheric pressures. However, for plants grown in hypobaria and high pCO2 (180 Pa) leaf damage was evident. Radish growth can be enhanced and transpiration reduced in hypobaria by enriching the gas phase with CO2 although at high levels leaf damage may occur.  相似文献

Loss of function of DNA repair genes has been implicated in the development of many types of cancer. In the last several years, heterozygosity leading to haploinsufficiency for proteins involved in DNA repair was shown to play a role in genomic instability and carcinogenesis after DNA damage is induced, for example by ionizing radiation. Since the effect of heterozygosity for one gene is relatively small, we hypothesize that predisposition to cancer could be a result of the additive effect of heterozygosity for two or more genes critical to pathways that control DNA damage signaling, repair or apoptosis. We investigated the role of heterozygosity for Atm, Rad9 and Brca1 on cell oncogenic transformation and cell survival induced by 1 GeV/n56Fe ions. Our results show that cells heterozygous for both Atm and Rad9 or Atm and Brca1 have high survival rates and are more sensitive to transformation by high energy iron ions when compared with wild-type controls or cells haploinsufficient for only one of these proteins. Since mutations or polymorphisms for similar genes exist in a small percentage of the human population, we have identified a radiosensitive sub-population. This finding has several implications. First, the existence of a radiosensitive sub-population may distort the shape of the dose–response relationship. Second, it would not be ethical to put exceptionally radiosensitive individuals into a setting where they may potentially be exposed to substantial doses of radiation.  相似文献

This study addresses the possibility of growing different halophytic plants on mineralized human urine as a way to recycle NaCl from human wastes in a bioregenerative life support system (BLSS). Two halophytic plant species were studied: the salt-accumulating Salicornia europaea and the salt-secreting Limonium gmelinii. During the first two weeks, plants were grown on Knop’s solution, then an average daily amount of urine produced by one human, which had been preliminarily mineralized, was gradually added to the experimental solutions. Nutrient solutions simulating urine mineral composition were gradually added to control solutions. NaCl concentrations in the stock solutions added to the experimental and control solutions were 9 g/L in the first treatment and 20 g/L in the second treatment. The mineralized human urine showed some inhibitory effects on S. europaea and L. gmelinii. The biomass yield of experimental plants was lower than that of control ones. If calculated for the same time period (120 d) and area (1 m2), the amount of sodium chloride taken up by S. europaea plants would be 11.7 times larger than the amount taken up by L. gmelinii plants (486 g/m2 vs. 41 g/m2). Thus, S. europaea is the better choice of halophyte for recycling sodium chloride from human wastes in BLSS.  相似文献

In the framework of satellite-only gravity field modeling, satellite laser ranging (SLR) data is typically exploited to recover long-wavelength features. This contribution provides a detailed discussion of the SLR component of GOCO02S, the latest release of combined models within the GOCO series. Over a period of five years (January 2006 to December 2010), observations to LAGEOS-1, LAGEOS-2, Ajisai, Stella, and Starlette were analyzed. We conducted a series of closed-loop simulations and found that estimating monthly sets of spherical harmonic coefficients beyond degree five leads to exceedingly ill-posed normal equation systems. Therefore, we adopted degree five as the spectral resolution for real data analysis. We compared our monthly coefficient estimates of degree two with SLR and Gravity Recovery and Climate Experiment (GRACE) time series provided by the Center for Space Research (CSR) at Austin, Texas. Significant deviations in C20 were noted between SLR and GRACE; the agreement is better for the non-zonal coefficients. Fitting sinusoids together with a linear trend to our C20 time series yielded a rate of (−1.75 ± 0.6) × 10−11/yr; this drift is equivalent to a geoid change from pole to equator of 0.35 ± 0.12 mm/yr or an apparent Greenland mass loss of 178.5 ± 61.2 km3/yr. The mean of all monthly solutions, averaged over the five-year period, served as input for the satellite-only model GOCO02S. The contribution of SLR to the combined gravity field model is highest for C20, and hence is essential for the determination of the Earth’s oblateness.  相似文献

Long-duration manned space missions mandate the development of a sustainable life support system and effective countermeasures against damaging space radiation. To mitigate the risk of inevitable exposure to space radiation, cultivation of fresh fruits and vegetables rich in antioxidants is an attractive alternative to pharmacological agents. However it has yet to be established whether antioxidant properties of crops can be preserved or enhanced in a space environment where environmental conditions differ from that which plants have acclimated to on earth. Scallion (Allium fistulosum) rich in antioxidant vitamins C and A, and flavonoids was used as a model plant to study the impact of a range of CO2 concentrations and light intensities that are likely encountered in a space habitat on food quality traits. Scallions were hydroponically grown in controlled environmental chambers under a combination of 3 CO2 concentrations of 400, 1200 and 4000 μmol mol−1 and 3 light intensity levels of 150, 300, 450 μmol m−2 s−1. Total antioxidant activity (TAA) of scallion extracts was determined using a radical cation scavenging assay. Both elevated CO2 and increasing light intensity enhanced biomass accumulation, but effects on TAA (based on dry weight) differed. TAA was reduced for plants grown under elevated CO2, but remained unchanged with increases in light intensity. Elevated CO2 stimulated greater biomass production than antioxidants, while an increase in photosynthetic photo flux promoted the synthesis of antioxidant compounds at a rate similar to that of biomass. Consequently light is a more effective stimulus than CO2 for antioxidant production.  相似文献

Growing plants to facilitate life in outer space, for example on the International Space Station (ISS) or at planned deep-space human outposts on the Moon or Mars, has received much attention with regard to NASA’s advanced life support system research. With the objective of in situ resource utilization to conserve energy and to limit transport costs, native materials mined on Moon or Mars are of primary interest for plant growth media in a future outpost, while terrestrial porous substrates with optimal growth media characteristics will be useful for onboard plant growth during space missions. Due to limited experimental opportunities and prohibitive costs, liquid and gas behavior in porous substrates under reduced gravity conditions has been less studied and hence remains poorly understood. Based on ground-based measurements, this study examined water retention, oxygen diffusivity and air permeability characteristics of six plant growth substrates for potential applications in space, including two terrestrial analogs for lunar and Martian soils and four particulate substrates widely used in reduced gravity experiments. To simulate reduced gravity water characteristics, the predictions for ground-based measurements (1 − g) were scaled to two reduced gravity conditions, Martian gravity (0.38 − g) and lunar gravity (0.16 − g), following the observations in previous reduced gravity studies. We described the observed gas diffusivity with a recently developed model combined with a new approach that estimates the gas percolation threshold based on the pore size distribution. The model successfully captured measured data for all investigated media and demonstrated the implications of the poorly-understood shift in gas percolation threshold with improved gas percolation in reduced gravity. Finally, using a substrate-structure parameter related to the gaseous phase, we adequately described the air permeability under reduced gravity conditions.  相似文献

The geometries, dipole moments, and rotational constants for the linear and/or bent cations, Cn+1H+ and CnN+(n = 1–6), were studied by the B3LYP method with the modest basis sets. For CnH+(n = odd; 3, 5, 7) and CnN+(n = even; 2, 4, 6), the theoretical rotational constants (Bes) of closed-shell singlet C3H+, C5H+, C7H+, CCN+, C4N+, and C6N+ were calculated to be about 11,244, 2420, 885.2, 11,970, 2439, and 880.8 MHz, respectively. By contrast, the triplets are stable than the corresponding singlets for CnH+(n = odd; 2, 4, 6) and CnN+(n = even; 3, 5) except CN+.  相似文献

A new set of data obtained at low solar activity from Ilorin, Nigeria (geog. latitude 8.5°N, geog longitude, 4.6°E, dip 4.1°S) is used to validate the IRI 2001 model at low solar activity. The results show in general a good agreement between model and observed B0 at night but an over estimation during daytime. The overestimation is greatest during the morning period (0600LT–1000LT). The model prediction for B1 is fairly good at night and during the day. A dependence of B0 on solar zenith angle χ is observed during the daytime. A formulation of the form B0 = A[cos(χ)n] is therefore proposed. Values of the constants n and A were determined for the period of low solar activity for this station.  相似文献

Euglena gracilis, a unicellular, photosynthetic flagellate is a model system for environmentally controlled behavior responses. The organism shows pronounced negative gravitaxis. This movement is based on physiological mechanisms, which in the past had been only indirectly assessed. It was shown that mechano-sensitive calcium channels are involved in the gravitaxis response. Recent studies have demonstrated that members of the transient receptor potential (TRP) family function as mechano-sensitive channels in several different cell types. We have sequenced part of a TRP gene in Euglena and applied RNA interference (RNAi) to confirm that these channels are involved in graviperception. It was found that RNAi against the putative TRP channel abolished gravitaxis. The genes of three calmodulins were sequences in Euglena, one of which was previously known in its protein structure (cal 1). The other two were unknown (cal 2 and cal 3). Cal 2 has been analyzed in detail. The biosynthesis of the corresponding proteins of cal 1 and cal 2 was inhibited by means of RNA interference to see whether this blockage impairs gravitaxis. RNAi of cal 1 leads to a long-term loss of free swimming in the cells (while euglenoid movement persists). It induced pronounced cell form aberrations and the division of cells was hampered. After recovery from RNAi the cell showed precise negative gravitaxis again. Thus cal 1 does not seem to be involved in gravitaxis. In contrast, the blockage of cal 2 has no pronounced influence on motility and cell form but leads to a complete loss of gravitactic orientation for more than 30 days showing that this calmodulin is an element in the signal transduction chain. The data are discussed in the context of the current model of the gravitaxis signal transduction chain in Euglena gracilis.  相似文献

We have used the technique of expansion in Empirical Orthogonal Functions (EOFs) to develop regional models of the critical frequencies of E and F2 layers (foE, foF2), peak height (hmF2), and semi-thickness of F2 layer (YmF2) over Pakistan. In the present study levels of solar activity specified by Smoothed Sunspot Number (R) from 10 to 200 are taken into account. The magnetic dip angle for the model ranges from 30° to 60°. We have compared the regional model and the International Reference Ionosphere (IRI) with measurements of three ionosondes in Pakistan. The model parameters foE and foF2 are found overall comparable to the observed hourly median values during daytime at Karachi (geographic latitude = 24.95°N, longitude = 67.13°E, magnetic inclination = 37°), Multan (30.18°N, 71.48°E, 45°) and Islamabad (33.75°N, 73.13°E, 51.5°) during the years 1988, 1996 and 2000. For hmF2 the computed values by regional and IRI model for the year 1995 are found close to each other. However, for YmF2the results are better during daytime as compared to nighttime.  相似文献

The spatial truncation error (STE) is a significant systematic error in the integral inversion of satellite gradiometric and orbital data to gravity anomalies at sea level. In order to reduce the effect of STE, a larger area than the desired one is considered in the inversion process, but the anomalies located in its central part are selected as the final results. The STE influences the variance of the results as well because the residual vector, which is contaminated with STE, is used for its estimation. The situation is even more complicated in variance component estimation because of its iterative nature. In this paper, we present a strategy to reduce the effect of STE on the a posteriori   variance factor and the variance components for inversion of satellite orbital and gradiometric data to gravity anomalies at sea level. The idea is to define two windowing matrices for reducing this error from the estimated residuals and anomalies. Our simulation studies over Fennoscandia show that the differences between the 0.5°×0.5°0.5°×0.5° gravity anomalies obtained from orbital data and an existing gravity model have standard deviation (STD) and root mean squared error (RMSE) of 10.9 and 12.1 mGal, respectively, and those obtained from gradiometric data have 7.9 and 10.1 in the same units. In the case that they are combined using windowed variance components the STD and RMSE become 6.1 and 8.4 mGal. Also, the mean value of the estimated RMSE after using the windowed variances is in agreement with the RMSE of the differences between the estimated anomalies and those obtained from the gravity model.  相似文献

The 53 kDa tumor suppressor protein p53 is generally thought to contribute to the genetic stability of cells and to protect cells from DNA damage through the activity of p53-centered signal transduction pathways. To clarify the effect of space radiation on the expression of p53-dependent regulated genes, gene expression profiles were compared between two human cultured lymphoblastoid cell lines: one line (TSCE5) has a wild-type p53 gene status, and the other line (WTK1) has a mutated p53 gene status. Frozen human lymphoblastoid cells were stored in a freezer in the International Space Station (ISS) for 133 days. Gene expression was analyzed using DNA chips after culturing the space samples for 6 h on the ground after their return from space. Ground control samples were also cultured for 6 h after being stored in a frozen state on the ground for the same time period that the frozen cells were in space. p53-Dependent gene expression was calculated from the ratio of the gene expression values in wild-type p53 cells and in mutated p53 cells. The expression of 50 p53-dependent genes was up-regulated, and the expression of 94 p53-dependent genes was down-regulated after spaceflight. These expression data identified genes which could be useful in advancing studies in basic space radiation biology. The biological meaning of these results is discussed from the aspect of gene functions in the up- and down-regulated genes after exposure to low doses of space radiation.  相似文献
