Aircraft parabolic flights provide repetitively up to 20 s of reduced gravity during ballistic flight manoeuvres. Parabolic flights are used to conduct short microgravity investigations in Physical and Life Sciences, to test instrumentation and to train astronauts before a space flight. The European Space Agency (ESA) has organized since 1984 thirty parabolic flight campaigns for microgravity research experiments utilizing six different airplanes. More than 360 experiments were successfully conducted during more than 2800 parabolas, representing a cumulated weightlessness time of 15 h 30 m. This paper presents the short duration microgravity research programme of ESA. The experiments conducted during these campaigns are summarized, and the different airplanes used by ESA are shortly presented. The technical capabilities of the Airbus A300 'Zero-G' are addressed. Some Physical Science, Technology and Life Science experiments performed during the last ESA campaigns with the Airbus A300 are presented to show the interest of this unique microgravity research tool to complement, support and prepare orbital microgravity investigations.  相似文献

Dating back to the Apollo and Skylab missions, it has been reported that astronauts suffered from bacterial and viral infections during space flight or after returning to Earth. Blood analyses revealed strongly reduced capability of human lymphocytes to become active upon mitogenic stimulation. Since then, a large number of in vitro studies on human immune cells have been conducted in space, in parabolic flights, and in ground-based facilities. It became obvious that microgravity affects cell morphology and important cellular functions. Observed changes include cell proliferation, the cytoskeleton, signal transduction and gene expression. This review gives an overview of the current knowledge of T cell regulation under altered gravity conditions obtained by in vitro studies with special emphasis on the cell culture conditions used. We propose that future in vitro experiments should follow rigorous standardized cell culture conditions, which allows better comparison of the results obtained in different flight- and ground-based experiment platforms.  相似文献

Free fall per se whether in parabolic or orbital flight may be regarded as a "partial" motion environment with respect to eliciting motion sickness, requiring an additional component to render this environment "complete" or stressful. Parabolic flight in toto falls in the category of a "complete" motion environment in that some persons became motion sick with head fixed and eyes closed. In the present experiment we selected subjects who were symptom free or nearly symptom free in the KC-135 with head fixed. All tests were conducted with the subject rotating at 30 rpm in a rotating litter chair, and comparisons were made between head-fixed and head-moving conditions (right-left) in the free-fall phase of parabolic flight and under simulated free-fall phases in the laboratory. With head fixed most subjects were insusceptible; with head moving left-right susceptibility was slightly higher in the laboratory than aloft. An additional comparison was made correlating susceptibility in the free-fall phases of parabolic flight with susceptibility to experimental motion sickness in Skylab. In both situations cross-coupled angular accelerations were generated by executing head and body movements out of the plane of rotation. In parabolic flight 9 of 15 subjects reached an endpoint just short of frank motion sickness. In the Skylab workshop all eight of the astronauts tested were symptom free at the end of the test. The explanation for the difference in susceptibility rests in two factors: (1) Basic susceptibility in free fall is lower than on the ground, and (2) in Skylab the astronauts who needed to adapt had achieved this goal prior to the first test on Mission-Day 8.  相似文献

The first ethological studies of astronauts' adaptation to microgravity dealt with the behavioral strategies observed during short-term space missions. No attempts had however been made to consider the initial moments of adaptation dynamics, when the subject is first submitted to conditions allowing body orientations in the full three dimensions of space. The present experimental approach was both longitudinal and transversal. It consisted of analysing, during a goal-directed orientation task in parabolic flight, the orientation behavior of 12 subjects with a past experience of 0, 30 or more than 300 parabolas. During each microgravity phase, the subjects were asked to orientate their bodies and touch, with the dominant hand, four coloured targets arranged inside the aircraft.

Results showed that for inexperienced subjects, the time between two target contacts was longer than experienced subjects. They often failed to reach all targets in the series during the first parabolas. They showed right-left confusion and a preference for the “up-down” vertical body orientation. Their performance, described by the efficiency of orientation in all three dimensions, improved over time and according to the level of experience. The results are discussed for the spontaneous, preliminary and integrative stages of adaptation, emphasizing new relationships between the body references and those of the surroundings. Such experiences lead the subject to develop a new mental representation of space.  相似文献

Physiological, morphological and biochemical studies of mammals flown onboard biosatellites of the series Cosmos revealed changes in their cardiovascular, musculoskeletal, endocrine and vestibular systems. Space flight resulted in moderate stress reactions, intralabyrinthine conflict information during movements and changes in fluid-electrolyte metabolism. Exposure to artificial gravity (1 g) decreased the level of myocardial, musculoskeletal and excretory changes, but disturbed the function of equilibrium. Studies with combined weightlessness and ionizing radiation demonstrated that weightlessness did not produce a significant modifying effect on radiation damage and postradiation recovery. Consistent changes in certain systems of animals and humans in weightlessness confirm the practical importance of biosatellite studies, which also contribute to the solution of general biology, problems associated with gravity effects on life processes.  相似文献

On Earth, the functional stretch reflex is an important component in the maintenance of posture and muscle tone. In parabolic flight experiments, we evaluated whether the functional stretch reflex, as reflected in the tonic vibration reflex, adjusts appropriately for changes in background gravitoinertial force level. Long-term sensitivity of human cells to reduced gravity has been supposed since the first Apollo missions and was demonstrated during several space missions in the past. However, little information is available on primary and rapid gravi-responsive elements in mammalian cells. In search of rapid-responsive molecular alterations in mammalian cells, short-term microgravity provided by parabolic flight maneuvers is an ideal way to elucidate such initial and primary effects. Modern biomedical research at the cellular and molecular level requires frequent repetition of experiments that are usually performed in sequences of experiments and analyses. Therefore, a research platform on Earth providing frequent, easy and repeated access to real microgravity for cell culture experiments is strongly desired. For this reason, we developed a research platform onboard the military fighter jet aircraft Northrop F-5E “Tiger II”. The experimental system consists of a programmable and automatically operated system composed of six individual experiment modules, placed in the front compartment, which work completely independent of the aircraft systems. Signal transduction pathways in cultured human cells can be investigated after the addition of an activator solution at the onset of microgravity and a fixative or lysis buffer after termination of microgravity. Before the beginning of a regular military training flight, a parabolic maneuver was executed. After a 1 g control phase, the parabolic maneuver starts at 13,000 ft and at Mach 0.99 airspeed, where a 22 s climb with an acceleration of 2.5g is initiated, following a free-fall ballistic Keplerian trajectory lasting 45 s with an apogee of 27,000 ft at Mach 0.4 airspeed. Temperature, pressure and acceleration are monitored constantly during the entire flight. Cells and activator solutions are kept at 37 °C during the entire experiment until the fixative has been added. The parabolic flight profile provides up to 45 s of microgravity at a quality of 0.05g in all axes. Access time is 30 min before take-off; retrieval time is 30 min after landing. We conclude that using military fighter jets for microgravity research is a valuable tool for frequent and repeated cell culture experiments and therefore for state-of-the art method of biomedical research.  相似文献

Dynamic ocular torsion was investigated in a group of healthy subjects during the course of parabolic flight by means of our video-based eye movement recording method-video-oculography. This technique enables a non-invasive dynamic measurement of all three dimensions of eye movement in a harsh experimental environment such as parabolic flight. The test subjects were positioned so that the changing resultant gravito-inertial field in the aircraft was aligned with their interaural (y) axis, primarily stimulating the utricular organs. The analysis of the torsional component of eye movement during the change of gravity between 1.8-0 and 0-1.8 g demonstrated a static component--well known as the ocular counter roll--and a dynamic component, which leads to a slight overshoot in the torsional response. These static and dynamic component of ocular torsion correlate with previous neurophysiological findings.  相似文献

This study identifies genes that determine survival during a space flight, using the model eukaryotic organism, Saccharomyces cerevisiae. Select strains of a haploid yeast deletion series grew during storage in distilled water in space, but not in ground based static or clinorotation controls. The survival advantages in space in distilled water include a 133-fold advantage for the deletion of PEX19, a chaperone and import receptor for newly- synthesized class I peroxisomal membrane proteins, to 77–40 fold for deletion strains lacking elements of aerobic respiration, isocitrate metabolism, and mitochondrial electron transport. Following automated addition of rich growth media, the space flight was associated with a marked survival advantage of strains with deletions in catalytically active genes including hydrolases, oxidoreductases and transferases. When compared to static controls, space flight was associated with a marked survival disadvantage of deletion strains lacking transporter, antioxidant and catalytic activity. This study identifies yeast deletion strains with a survival advantage during storage in distilled water and space flight, and amplifies our understanding of the genes critical for survival in space.  相似文献

Aircraft parabolic flights provide repetitively short periods of reduced gravity by flying a ballistic trajectory, preceded and followed by periods of acceleration up to 2g. The ballistic part has a shape of an arc of a parabola and is quite commonly referred to as a parabolic flight.In this short note, we want to show that, although the wording parabolic is stricto sensu incorrect, it is though not far from reality and we show how to calculate simply the approximation committed by calling this trajectory parabolic.  相似文献

In plants, sensitive and selective mechanisms have evolved to perceive and respond to light and gravity. We investigated the effects of microgravity on the growth and development of Arabidopsis thaliana (ecotype Landsberg) in a spaceflight experiment. These studies were performed with the Biological Research in Canisters (BRIC) hardware system in the middeck region of the space shuttle during mission STS-131 in April 2010. Seedlings were grown on nutrient agar in Petri dishes in BRIC hardware under dark conditions and then fixed in flight with paraformaldehyde, glutaraldehyde, or RNAlater. Although the long-term objective was to study the role of the actin cytoskeleton in gravity perception, in this article we focus on the analysis of morphology of seedlings that developed in microgravity. While previous spaceflight studies noted deleterious morphological effects due to the accumulation of ethylene gas, no such effects were observed in seedlings grown with the BRIC system. Seed germination was 89% in the spaceflight experiment and 91% in the ground control, and seedlings grew equally well in both conditions. However, roots of space-grown seedlings exhibited a significant difference (compared to the ground controls) in overall growth patterns in that they skewed to one direction. In addition, a greater number of adventitious roots formed from the axis of the hypocotyls in the flight-grown plants. Our hypothesis is that an endogenous response in plants causes the roots to skew and that this default growth response is largely masked by the normal 1?g conditions on Earth.  相似文献

In the period from March 2003 to April 2005 we fulfilled five experimental cultivations of genetically marked dwarf pea species in greenhouse Lada installed in the Russian segment (RS) of the International Space Station (ISS). The purpose of this series of experiments was to make morphologic and genetic analysis of pea plants grown in successive generations.According to our results, pea growth and development over the full cycle of ontogenesis (from seed to seed) taking place in space greenhouse Lada were not different as compared with the ground control plants. In addition, four successive pea crops gathered in space flight did not loose their reproductive functions and formed viable seeds.Genetic analysis of the plants grown from the “space” and “ground” seeds produced by the first to fourth successive crops was performed using the methods of chromosomal aberrations count and Random Amplified Polymorphic DNA (molecular method). No genetic polymorphism was found either in the experimental or control crops. This can serve as a sound argument for the supposition that the genetic apparatus of plants is not impacted by exposure of several successive generations to the conditions of space flight.  相似文献

The activity of the enzymes involved in aminoacid metabolism (tyrosine aminotransferase, TAT, tryptophan pyrrolase TP, serine dehydratase, SD) with rapid response to glucocorticoids and enzymes requiring for activity increase repeated administration of corticosterone (alanine aminotransferase, ALT, aspartate aminotransferase, AST) in liver, the changes of lipolysis in adipose tissue and the plasma corticosterone levels were studied in rats subjected to space flight (F), in animals from synchron model experiments (SM, simulated conditions of space flight in laboratory) and in intact controls (C). The increase of plasma corticosterone concentration and of the activity of rapidly (TAT, TP, SD) and slowly activating enzymes (ALT, AST) was found in F group 6-10 hr after space flight (18.5 days on biosatellite COSMOS 1129). This suggested the presence of acute-stress (associated primarily with the landing) and chronic stress induced hypercorticosteronemia during the flight. After the short 6-day period of recovery the plasma corticosterone concentrations and the activities of liver enzymes returned to control levels. The exposition of animals to repeated immobilization stress showed higher response of corticosterone levels in flight rats as compared to intact controls. No changes in basal lipolysis were observed in flight rats in comparison to intact controls, however the stimulation of lipolysis by norepinephrine was lower in animals from F and SM groups. This lower response of lipolytic processes to norepinephrine was found in flight animals also after six days period of recovery. These results showed that there are important changes in the regulation of lipolytic processes in adipose tissue of rats after space flight and in the conditions of model experiments.  相似文献

The aim of this work was to analyze the possible alteration of thyrotropin (TSH) receptors in microgravity, which could explain the absence of thyroid cell proliferation in the space environment. Several forms of the TSH receptor are localized on the plasma membrane associated with caveolae and lipid rafts. The TSH regulates the fluidity of the cell membrane and the presence of its receptors in microdomains that are rich in sphingomyelin and cholesterol. TSH also stimulates cyclic adenosine monophosphate (cAMP) accumulation and cell proliferation. Reported here are the results of an experiment in which the FRTL-5 thyroid cell line was exposed to microgravity during the Texus-44 mission (launched February 7, 2008, from Kiruna, Sweden). When the parabolic flight brought the sounding rocket to an altitude of 264?km, the culture media were injected with or without TSH in the different samples, and weightlessness prevailed on board for 6 minutes and 19 seconds. Control experiments were performed, in parallel, in an onboard 1g centrifuge and on the ground in Kiruna laboratory. Cell morphology and function were analyzed. Results show that in microgravity conditions the cells do not respond to TSH treatment and present an irregular shape with condensed chromatin, a modification of the cell membrane with shedding of the TSH receptor in the culture medium, and an increase of sphingomyelin-synthase and Bax proteins. It is possible that real microgravity induces a rearrangement of specific sections of the cell membrane, which act as platforms for molecular receptors, thus influencing thyroid cell function in astronauts during space missions.  相似文献

Commercial suborbital space flights will reach altitudes above 100 km, with 3–5 min of weightlessness bracketed by high-g launch and landing phases. The proposed frequency of these flights, and the large passenger population, present interesting opportunities for researchers in the life sciences. The characteristics of suborbital flight are between those of parabolic and orbital flights, opening up new scientific possibilities and easing the burden for obtaining access to 0g.  相似文献

Based on systematic studies of the impact of space flight factors on higher plants, carried out for many years both aboard orbital research stations and in the ground laboratories, we have formulated the means of solving the problem of the interaction of mankind with the plant life. The principal feasibility of the growth and development of higher plants (arabidopsis, peas, and wheat) under conditions of space flight is established, including the generative stage (arabidopsis). It is proved that the factors of space flight destabilize the macro- and microstructures of higher plants and their cellular organelles, which is an unambiguous response of cells to the changing conditions of their existence.  相似文献

When discussing problems related to medical service in space flight, particular attention should be given to the specific living conditions and changes associated with space flight. In disease and injury, surgery can be provided only after conservative therapy has failed. In this context gnotobiological chambers allowing surgery in aseptic conditions seem promising. A portable set of interchangeable surgical tools should be made of light-weight alloys that can be readily sterilized. Electroanalgesia in combination with auriculoacupuncture as well as peridureal anesthesia may be used as they allow normal operations in autonomous space flight conditions. Changes in the sympatho-adrenal and kallikrein-kinin systems, as well as water-electrolyte balance, should be taken into account in developing methods and means of medical service in critical situations. Special attention should be given to the prevention and treatment of brain edema in view of weightlessness-induced cephalad fluid shifts.  相似文献
