The space radiation environment is a mixed field consisting of different particles having different energies, including high charge and energy (HZE) ions. Conventional measurements of absorbed doses may not be sufficient to completely characterise the radiation field and perform reliable estimates of health risks. Biological dosimetry, based on the observation of specific radiation-induced endpoints (typically chromosome aberrations), can be a helpful approach in case of monitored exposure to space radiation or other mixed fields, as well as in case of accidental exposure. Furthermore, various ratios of aberrations (e.g. dicentric chromosomes to centric rings and complex exchanges to simple exchanges) have been suggested as possible fingerprints of radiation quality, although all of them have been subjected to some criticisms. In this context a mechanistic model and a Monte Carlo code for the simulation of chromosome aberration induction were developed. The model, able to provide dose-responses for different aberrations (e.g. dicentrics, rings, fragments, translocations, insertions and other complex exchanges), was further developed to assess the dependence of various ratios of aberrations on radiation quality. The predictions of the model were compared with available data, whose experimental conditions were faithfully reproduced. Particular attention was devoted to the scoring criteria adopted in different laboratories and to possible biases introduced by interphase death and mitotic delay. Understanding the effects of single-particles from conventional radiation biology experiments is problematic due to the stochastics of particle tracks. This complicates the determinations of risk associated with low doses. We have developed a charged particle microbeam, which allows individually counted particles to be delivered to precise cellular locations. The system is capable of delivering a single charged particle with > 99% efficiency. Of these particles 90% are delivered with a resolution of +/- 2 micrometers and 96% with a resolution of +/- 5 micrometers. We have carried out preliminary studies in Chinese hamster V79 cells to monitor the effectiveness of low energy protons at inducing cytological damage. We have used the micronucleus assay as a measure of predominantly lethal chromosome damage. The effects of a single 3.2 MeV proton delivered individually to cells could be measured, with less than 2% of the exposed cells producing micronuclei 24 hours later. The yield of micronuclei formation was essentially linear up to the highest dose (30 particles per cell nucleus) delivered. Ultimately, the ability to target particles to different parts of the cell nucleus may start to impact on models available for chromosome aberration formation and chromosomal Organisation and mechanisms underlying genomic instability.  相似文献

The cytogenetic effects of X-rays and Au ions were investigated in repair-proficient CHO-K1 cells and their radiosensitive mutant strain xrs5, which shows a defect in the rejoining of DNA double-strand breaks. Both cell lines were synchronized by mitotic shake off, irradiated in G1-phase with either 250 kV X-rays or 780 MeV/u Au ions (LET: 1150 keV/micrometer) and chromosome aberrations were analyzed in first post-irradiation metaphases. Isoeffective doses of X-rays for the induction of aberrant cells and aberrations per cell were about 14 times lower for xrs5 than for CHO-K1 cells. After high LET radiation the difference in the cytogenetic response of both cell lines was drastically diminished. Furthermore, the analysis of the aberration types induced by sparsely and densely ionizing radiation showed for both cell lines specific changes in the spectrum of aberration types as LET increases. The experimental results are discussed with respect to the different types of lesions induced by sparsely and densely ionizing radiation.  相似文献

Heavy ions are more efficient in producing complex-type chromosome exchanges than sparsely ionizing radiation, and this can potentially be used as a biomarker of radiation quality. We measured the induction of complex-type chromosomal aberrations in human peripheral blood lymphocytes exposed in vitro to accelerated H-, He-, C-, Ar-, Fe- and Au-ions in the LET range of approximately 0.4-1400 keV/micrometers. Chromosomes were analyzed either at the first post-irradiation mitosis, or in interphase, following premature condensation by phosphatase inhibitors. Selected chromosomes were then visualized after FISH-painting. The dose-response curve for the induction of complex-type exchanges by heavy ions was linear in the dose-range 0.2-1.5 Gy, while gamma-rays did not produce a significant increase in the yield of complex rearrangements in this dose range. The yield of complex aberrations after 1 Gy of heavy ions increased up to an LET around 100 keV/micrometers, and then declined at higher LET values. When mitotic cells were analyzed, the frequency of complex rearrangements after 1 Gy was about 10 times higher for Ar- or Fe- ions (the most effective ions, with LET around 100 keV/micrometers) than for 250 MeV protons, and values were about 35 times higher in prematurely condensed chromosomes. These results suggest that complex rearrangements may be detected in astronauts' blood lymphocytes after long-term space flight, because crews are exposed to HZE particles from galactic cosmic radiation. However, in a cytogenetic study of ten astronauts after long-term missions on the Mir or International Space Station, we found a very low frequency of complex rearrangements, and a significant post-flight increase was detected in only one out of the ten crewmembers. It appears that the use of complex-type exchanges as biomarker of radiation quality in vivo after low-dose chronic exposure in mixed radiation fields is hampered by statistical uncertainties.  相似文献

A broad spectrum of particles and energies has been used in the last years to study the influence of the radiation quality i.e. of the physical parameters of the particle beam on the biological effectiveness ?2?12?. In these measurements a common structure of the functional dependence of the induction probability per particle (cross section) from the linear energy transfer is observed for different biological endpoints. Because of the relevance for space research, we present in this report our data from experiments with iron and nickel particles, in particular. Our experiments were designed to investigate the relationship between the inactivation and chromosome aberration in mammalian cells and the induction of single and double strand breaks in SV40 DNA in respect to the parameters of the track formation like LET and particle energy.  相似文献

Chromosomal intrachanges induced by swift iron ions.   总被引:1,自引:0,他引:1  
We measured the induction of structural aberrations in human chromosome 5 induced by iron ions using the novel technique of multicolor banding in situ hybridization (mBAND). Human lymphocytes isolated from whole blood were exposed in vitro to 500 MeV/n (LET=200 keV/micrometers, doses 1 or 4 Gy) Fe nuclei at the HIMAC accelerator in Chiba (Japan). Chromosomes were prematurely condensed by calyculin A after 48 h in culture and slides were painted by mBAND. We found a frequency of 0.11 and 0.57 residual breakpoints per chromosome 5 after 1 and 4 Gy Fe-ions, respectively. Inter-chromosomal exchanges were the prevalent aberration type measured at both doses, followed by terminal deletions, and by intra-chromosomal exchanges. Among intra-chromosomal exchanges, intra-arm events were more frequent than inter-arm, but a significant number of intra-changes was associated to inter-changes involving the same chromosome after 4 Gy of iron ions. These events show that the complexity of chromosomal exchanges induced by heavy ions can be higher than expected by previous FISH studies.  相似文献

Gene mutations can be induced by radiation as a result of chromosomal translocations. A biophysical model is developed to estimate the frequency of this type of mutation induced by low-LET radiation. Mutations resulting from translocations are assumed to be formed by misrejoining of two DNA double strand breaks (DSB), one within the gene and one on a different chromosome. The chromosome containing the gene is assumed to occupy a spherical territory and does not overlap spatially with other chromosomes. Misrejoining between two DSB can occur only if the two DSB are closer than an interaction distance at the time of their induction. Applying the model to mutations of the hprt gene induced in G0 human lymphocyte cells by low-LET radiation, it is calculated that mutations resulting from translocations account for about 14% of the total mutations. The value of the interaction distance is determined to be 0.6 micrometers by comparing with the observed frequency of translocations in the X-chromosome.  相似文献

Purpose

To evaluate radiation induced chromosome aberration frequency in peripheral blood lymphocytes of cosmonauts who participated in flights on Mir Orbital Station and ISS (International Space Station).

Materials and methods

Cytogenetic examination which has been performed in the period 1992–2008 included the analysis of chromosome aberrations using conventional Giemsa staining method in 202 blood samples from 48 cosmonauts who participated in flights on Mir Orbital Station and ISS.

Results

Space flights led to an increase of chromosome aberration frequency. Frequency of dicentrics plus centric rings (Dic+Rc) depend on the space flight duration and accumulated dose value. After the change of space stations (from Mir Orbital Station to ISS) the radiation load of cosmonauts based on data of cytogenetic examination decreased. Extravehicular activity also adds to chromosome aberration frequency in cosmonauts’ blood lymphocytes. Average doses after the first flight, estimated by the frequency of Dic+Rc, were 227 and 113 mGy Eq for long-term flights (LTF) and 107 and 53 mGy Eq for short-term flights (STF).

Conclusion

Cytogenetic examination of cosmonauts can be applied to assess equivalent doses.  相似文献

Biological damages such as mutations, chromosomal aberrations etc. are a consequence of biochemical changes mostly in the DNA. With ionizing radiation, these chemical changes are due to primary ionization events and secondary ionization effects caused by the primarily produced electrons. Differences in the biological response of densely ionizing radiation, like heavy charged particles, in comparison to sparsely ionizing radiation, such as X- or gamma-rays, are mainly due to the differences in the production of the so called delta-electrons. Therefore, the emission process of electrons i.e. the cross section for the primary ionization event as well as the energy and angular distribution of the emitted electrons should be understood in detail. The delta-electron emission processes occuring in fast heavy ion atom collisions are explained qualitatively. The different spectral structures of electron emission arising from either the target or the projectile are explained in terms of simple models of the kinetics of momentum transfer induced by the COULOMB forces. In collisions of very heavy ions with matter, high nuclear COULOMB forces are created. These forces lead to a strong polarization of the electronic states of the participated electrons. The effects of this polarization are discussed.  相似文献

针对突防目标不同角域内RCS(Radar Cross Section)差异较大的情况,提出5种不同的突防目标特征信号雷达重点探测区域建模方案.在计算机仿真平台上,根据目标的周向散射特性,按照这5种不同的模型取RCS均值计算雷达对目标的探测概率值.不同建模方案所得探测概率值与精确探测概率值的平均误差分别为: 13.66%,12.35%,6.6%,5.07%和2.4%.平均误差的差异表明:采用5个或8个特征信号重点探测区域的建模方案,所得探测概率值的误差小,能反映出雷达对目标探测概率的动态变化过程.仿真平台中,这两种建模方案无需存储大量目标的RCS值,只需记录5个或8个目标的RCS均值,可节省计算机的存储量,提高计算速度.  相似文献

For the evaluation of organ dose and dose equivalent of astronauts on space shuttle and the International Space Station (ISS) missions, the CAMERA models of CAM (Computerized Anatomical Male) and CAF (Computerized Anatomical Female) of human tissue shielding have been implemented and used in radiation transport model calculations at NASA. One of new human geometry models to meet the “reference person” of International Commission on Radiological Protection (ICRP) is based on detailed Voxel (volumetric and pixel) phantom models denoted for male and female as MAX (Male Adult voXel) and FAX (Female Adult voXel), respectively. We compared the CAM model predictions of organ doses to those of MAX model, since the MAX model represents the male adult body with much higher fidelity than the CAM model currently used at NASA. Directional body-shielding mass was evaluated for over 1500 target points of MAX for specified organs considered to be sensitive to the induction of stochastic effects. Radiation exposures to solar particle event (SPE), trapped protons, and galactic cosmic ray (GCR) were assessed at the specific sites in the MAX phantom by coupling space radiation transport models with the relevant body-shielding mass. The development of multiple-point body-shielding distributions at each organ made it possible to estimate the mean and variance of organ doses at the specific organ. For the estimate of doses to the blood forming organs (BFOs), data on active marrow distributions in adult were used to weight the bone marrow sites over the human body. The discrete number of target points of MAX organs resulted in a reduced organ dose and dose equivalent compared to the results of CAM organs especially for SPE, and should be further investigated. Differences of effective doses between the two approaches were found to be small (<5%) for GCR.  相似文献

In order to understand the effects of shielding on the induction of biological damages by charged particles, we conducted experiments with accelerated protons (250 MeV) and iron particles (1 GeV/u). Human lymphocytes in vitro were exposed to particle beams through polyethylene with various thickness, and chromosomal aberrations were determined using FISH technique. Dose response curves for chromosome aberrations were obtained and compared for various particle types. Experimental results indicated that for a given absorbed dose at the cell, the effectiveness of protons and iron particles in the induction of chromosomal aberrations was not significantly altered by polyethylene with thickness up to 30-cm and 15-cm respectively. Comparing with gamma rays, charged particles were very effective in producing complex chromosomal damages, which may be an important mechanism in alterating functions in nondividing tissues, such as nervous systems.  相似文献

Karyotypes of human cells surviving X- and alpha-irradiation have been studied. Human mammary epithelial cells of the immortal, non-tumorigenic cell line H184B5 F5-1 M/10 were irradiated and surviving clones isolated and expanded in culture. Cytogenetic analysis was performed using dedicated software with an image analyzer. We have found that both high- and low-LET radiation induced chromosomal instability in long-term cultures, but with different characteristics. Complex chromosomal rearrangements were observed after X-rays, while chromosome loss predominated after alpha-particles. Deletions were observed in both cases. In clones derived from cells exposed to alpha-particles, some cells showed extensive chromosome breaking and double minutes. Genomic instability was correlated to delayed reproductive death and neoplastic transformation. These results indicate that chromosomal instability is a radiation-quality-dependent effect which could determine late genetic effects, and should therefore be carefully considered in the evaluation of risk for space missions.  相似文献

It has been known for many years that relationships between absorbed dose and biological effect vary with the type of radiation. In particular, neutrons and alpha particles are more damaging than x or gamma radiations. This applies to a range of biological effects such as cell killing, chromosome aberrations, cell mutation, cell transformation as well as life shortening and cancer induction in animals. The application of this knowledge to devise a scheme for specifying the quality factor (Q) in radiological protection has been the subject of much debate. There are no tumour data in humans from which the quality factor may be derived. The problems of using animal and cell transformation data which are probably the next best choice are discussed. The extensive data base on chromosomal aberrations in human lymphocytes is described and discussed in terms of relevance to deducing quality factors. Particular emphasis is placed on data obtained at low doses and low dose rates.  相似文献

The induction of chromosome aberrations by heavy charged particles was studied in V79 Chinese hamster cells over a wide range of energies (3-100 MeV/u) and LET (20-16000 keV/micrometer). For comparison, X-ray experiments were performed. Our data indicate quantitative and qualitative differences in the response of cells to particle and x-ray irradiation. For the same level of cell survival the amount of damaged cells which can be observed is smaller in heavy ion (11.4 MeV/u Ar) irradiated samples. The highest yield of damaged cells is found 8 to 12 hours after particle irradiation and 4 hours after x-irradiation. Differences in the amount of damaged cells are attributed to cell cycle perturbations which interfere with the expression of damage. After heavy ion exposure the amount of cells reaching mitosis (mitotic index) decreases drastically and not all damaged cells reach mitosis within 48 hours after exposure. A portion of cells die in interphase. Cell cycle delays induced by x-ray irradiation are less pronounced and all cells reach the first post-irradiation mitosis within 24 hours after irradiation. Additionally, the damage produced by charged particles seems to be more severe. The disintegration of chromosomes was only observed after high LET radiation: an indication of the high and local energy deposition in the particle track. Only cross sections for the induction of chromosome aberrations in mitotic cells were reported in this paper because of the problems arising from the drastic cell cycle perturbations. In this case, cells were irradiated in mitosis and assayed immediately.  相似文献

We present measurements of LET spectra for near earth orbits with various inclinations and altitudes. A comparison with calculated LET spectra shows that the contribution from direct ionizing galactic cosmic rays is well described by the models. An additional contribution to the spectra originates from stopping protons and from nuclear interactions of particles with material. In the case of an interaction a large amount of energy is deposited in a small volume by target recoils or target fragments. These events will be called short range (SR) events. For a low inclination orbit radiation belt protons are the main source of these events while galactic protons become more important when increasing the inclination to near polar orbits. We show that the contribution of SR events for orbits with low altitude (324 km) and 57 degrees inclination is comparable to that for an orbit with 28 degrees inclination at a high altitude (510 km).  相似文献

It has been known for some time that adequate assessment of spacecraft shield requirements and concomitant estimates of astronauts radiation exposures from galactic cosmic radiation requires accurate, quantitative methods for characterizing these radiation fields as they pass through thick absorbers. The main nuclear interaction processes involved are (1) nuclear elastic and inelastic collisions, and (2) nuclear breakup (fragmentation) and electromagnetic dissociation (EMD). Nuclear fragmentation and EMD are important because they alter the elemental and isotopic composition of the transported radiation fields. At present, there is no suitably accurate theory for predicting nuclear fragmentation cross sections for all collision pairs and energies of interest in space radiation protection. Typical cross-section differences between theory and experiment range from about 25 percent to a factor of two. The resulting errors in transported flux, for high linear energy transfer (LET) particles, are comparble to these cross-section errors. In this overview, theoretical models of heavy ion fragmentation currently used to generate input data bases for cosmic-ray transport and shielding codes are reviewed. Their shortcomings are discussed. Further actions needed to improve their accuracy and generality are presented.  相似文献

A survey of empirical models of particles (electrons, protons and heavier ions) of the Earth's radiation belts developed to date is presented. Results of intercomparison of the different models as well as comparison with experimental data are reported.
