Bacterial spores have been considered as microbial life that could survive interplanetary transport by natural impact processes or human spaceflight activity. Deposition of terrestrial microbes or their biosignature molecules onto the surface of Mars could negatively impact life detection experiments and planetary protection measures. Simulated Mars solar radiation, particularly the ultraviolet component, has been shown to reduce spore viability, but its effect on spore germination and resulting production of biosignature molecules has not been explored. We examined the survival and germinability of Bacillus subtilis spores exposed to simulated martian conditions that include solar radiation. Spores of B. subtilis that contain luciferase resulting from expression of an sspB-luxAB gene fusion were deposited on aluminum coupons to simulate deposition on spacecraft surfaces and exposed to simulated Mars atmosphere and solar radiation. The equivalent of 42 min of simulated Mars solar radiation exposure reduced spore viability by nearly 3 logs, while germination-induced bioluminescence, a measure of germination metabolism, was reduced by less than 1 log. The data indicate that spores can retain the potential to initiate germination-associated metabolic processes and produce biological signature molecules after being rendered nonviable by exposure to Mars solar radiation. 相似文献

Bacterial endospores in the genus Bacillus are considered good models for studying interplanetary transfer of microbes by natural or human processes. Although spore survival during transfer itself has been the subject of considerable study, the fate of spores in extraterrestrial environments has received less attention. In this report we subjected spores of a strain of Bacillus subtilis, containing luciferase resulting from expression of an sspB-luxAB gene fusion, to simulated martian atmospheric pressure (7-18 mbar) and composition (100% CO(2)) for up to 19 days in a Mars simulation chamber. We report here that survival was similar between spores exposed to Earth conditions and spores exposed up to 19 days to simulated martian conditions. However, germination-induced bioluminescence was lower in spores exposed to simulated martian atmosphere, which suggests sublethal impairment of some endogenous spore germination processes. 相似文献

One design for a manned Mars base incorporates a bioregenerative life support system based upon growing higher plants at a low atmospheric pressure in a greenhouse on the Martian surface. To determine the concept's feasibility, the germination and initial growth of wheat (Triticum aestivum) was evaluated at low atmospheric pressures in simulated Martian atmosphere (SMA) and in SMA supplemented with oxygen. Total atmospheric pressures ranged from 10 to 1013 mb. No seeds germinated in pure SMA, regardless of atmospheric pressure. In SMA plus oxygen at 60 mb total pressure, germination and growth occurred but were lower than in the Earth atmosphere controls. 相似文献

We have been studying the survival rates of some species of terrestrial unicellular and multicellular organism (viruses, bacteria, yeasts, fungi, algae, etc.) under simulated interstellar conditions, in connection with planetary quarantine. The interstellar environment in the solar system has been simulated by low temperature, high vacuum (77 K, 4 x 10(-8) torr), and proton irradiation from a Van de Graaff generator. After exposure to a barrage of protons corresponding to about 250 years of irradiation in solar space, tobacco mosaic virus, Bacillus subtilis spores, Staphylococcus aureus, Micrococcus flavus, Aspergillus niger spores, and Clostridium mangenoti spores showed survival rates of 82, 45, 74, 13, 28, and 25%, respectively. 相似文献

To prevent forward contamination and maintain the scientific integrity of future life-detection missions, it is important to characterize and attempt to eliminate terrestrial microorganisms associated with exploratory spacecraft and landing vehicles. Among the organisms isolated from spacecraft-associated surfaces, spores of Bacillus pumilus SAFR-032 exhibited unusually high resistance to decontamination techniques such as UV radiation and peroxide treatment. Subsequently, B. pumilus SAFR-032 was flown to the International Space Station (ISS) and exposed to a variety of space conditions via the European Technology Exposure Facility (EuTEF). After 18 months of exposure in the EXPOSE facility of the European Space Agency (ESA) on EuTEF under dark space conditions, SAFR-032 spores showed 10-40% survivability, whereas a survival rate of 85-100% was observed when these spores were kept aboard the ISS under dark simulated martian atmospheric conditions. In contrast, when UV (>110?nm) was applied on SAFR-032 spores for the same time period and under the same conditions used in EXPOSE, a ~7-log reduction in viability was observed. A parallel experiment was conducted on Earth with identical samples under simulated space conditions. Spores exposed to ground simulations showed less of a reduction in viability when compared with the "real space" exposed spores (~3-log reduction in viability for "UV-Mars," and ~4-log reduction in viability for "UV-Space"). A comparative proteomics analysis indicated that proteins conferring resistant traits (superoxide dismutase) were present in higher concentration in space-exposed spores when compared to controls. Also, the first-generation cells and spores derived from space-exposed samples exhibited elevated UVC resistance when compared with their ground control counterparts. The data generated are important for calculating the probability and mechanisms of microbial survival in space conditions and assessing microbial contaminants as risks for forward contamination and in situ life detection. 相似文献

“Mars-105” experiment was executed in March–July 2009 in Moscow, at the Institute for Bio-Medical Problems (IBMP) with participation of European Space Agency (ESA) to simulate some specific conditions of future piloted Mars mission. In the last 35 days of isolation, in order to simulate autonomous flight conditions, some serious restrictions were established for the crew resupply and communication with Mission Control (MC). The objective of the study was to investigate psychophysiological and behavioral aspects (communication) of adaptation during this period of “high autonomy”. We used computerized analysis of the crew written daily reports to calculate the frequencies of utilization of certain semantic units, expressing different psychological functions. To estimate the level of psycho-physiological stress, we measured the concentration of urinal cortisol once in two weeks. To investigate psycho-emotional state, we used the questionnaire SAN, estimating Mood, Activity and Health once in two weeks.During the simulation of autonomous flight, we found out the different tendencies of communicative behavior. One group of subjects demonstrated the tendency to “activation and self-government” under “high autonomy” conditions. The other subjects continued to use communicative strategy that we called “closing the communication channel”. “Active” communication strategy was accompanied by increasing in subjective scores of mood and activity. The subjects, whose communication strategy was attributed as “closing”, demonstrated the considerably lower subjective scores of mood and activity. Period of high autonomy causes specific changes in communication strategies of the isolated crew. 相似文献

In the present investigation we report the effects of simulated microgravity conditions (clinostat) on the induction of chromosomal aberrations in human lymphocytes in vitro by (R) Bleomycin. Chromosomal aberrations have been analysed by means of fluorescent in situ hybridisation (FISH) and chromosome-specific composite DNA probes (chromosome painting). The results obtained show that, under simulated microgravity conditions, the levels of both symmetrical and asymmetrical (dicentrics, rings), the number of cells bearing "complex" aberrations and hence the total numbers of aberrations were significantly elevated at any of the dose-levels assayed, compared to the parallel treatments performed as 1g control ("ground"). Furthermore, the ratio symmetrical:asymmetrical translocations was markedly elevated under simulated microgravity conditions, compared to the findings usually observed under "normal" 1g conditions. On these bases, we are much inclined to believe that simulated microgravity, rather than limiting the resealing of DNA double strand breaks (DSB's) induced by genotoxic agents is influencing in terms of enhancement the misrejoining of DSB's which is actually responsible for the fixation of the original lesions to DNA into chromosomal aberrations. In addition, the possible different misrepair processes leading to the formation of symmetrical and asymmetrical translocations might be differentially influenced by microgravity being the symmetrical translocations significantly more represented. 相似文献

Viability rates were determined for microbial populations of Escherichia coli and Deinococcus radiodurans under the environmental stresses of low temperature (-35 degrees C), low-pressure conditions (83.3 kPa), and ultraviolet (UV) irradiation (37 W/m(2)). During the stress tests the organisms were suspended in saltwater soil and freshwater soil media, at variable burial depths, and in seawater. Microbial populations of both organisms were most susceptible to dehydration stress associated with low-pressure conditions, and to UV irradiation. However, suspension in a liquid water medium and burial at larger depths (5 cm) improved survival rates markedly. Our results indicate that planetary surfaces that possess little to no atmosphere and have low water availability do not constitute a favorable environment for terrestrial microorganisms. 相似文献

Bacterial spores have been used as model systems for studying the theory of interplanetary transport of life by natural processes such as asteroidal or cometary impacts (i.e., lithopanspermia). Because current spallation theory predicts that near-surface rocks are ideal candidates for planetary ejection and surface basalts are widely distributed throughout the rocky planets, we isolated spore-forming bacteria from the interior of near-subsurface basalt rocks collected in the Sonoran desert near Tucson, Arizona. Spores were found to inhabit basalt at very low concentrations (=28 colony-forming units/g) in these samples. Six isolates identified as being most closely related to Bacillus pumilus and one Bacillus subtilis isolate were recovered from near-subsurface basalt samples. Populations of purified spores prepared from the isolated strains were subjected to 254-nm UV and ballistics tests in order to assess their resistance to UV radiation and to extreme acceleration shock, two proposed lethal factors for spores during interplanetary transfer. Specific natural isolates of B. pumilus were found to be substantially more resistant to UV and extreme acceleration than were reference laboratory strains of B. subtilis, the benchmark organism, suggesting that spores of environmental B. pumilus isolates may be more likely to survive the rigors of interplanetary transfer. 相似文献

It was previously shown that different cell types in vivo and in vitro may die via apoptosis under weightlessness conditions in space as well as in simulated hypogravity on the Earth. We assessed survivability of spinal motoneurons of rats after 35-day antiorthostatic hind limb suspension. Following weight bearing, unloading the total protein content in lumbar spinal cord is dropped by 21%. The electrophysiological studies of m. gastrocnemius revealed an elevated motoneurons’ reflex excitability and conduction disturbances in the sciatic nerve axons. The number of myelinated fibers in the ventral root of experimental animals was insignificantly increased by 35-day of antiorthostatic hind limb suspension, although the retrograde axonal transport was significantly decreased during the first week of simulated hypogravity. The results of the immunohistochemical assay with antibodies against proapoptotic protein caspase 9 and cytotoxicity marker neuron specific nitric oxide synthase (nNOS) and the TUNEL staining did not reveal any signs of apoptosis in motoneurons of suspended and control animals. To examine the possible adaptation mechanisms activated in motoneurons in response to simulated hypogravity we investigated immunoexpression of Hsp25 and Hsp70 in lumbar spinal cord of the rats after 35-day antiorthostatic hind limb suspension. Comparative analysis of the immunohistochemical reaction with anti-Hsp25 antibodies revealed differential staining of motoneurons in intact and experimental animals. The density of immunoprecipitate with anti-Hsp25 antibodies was substantially higher in motoneurons of the 35-day suspended than control rats and the more intensive precipitate in this reaction was observed in motoneuron neuritis. Quantitative analysis of Hsp25 expression demonstrated an increase in the Hsp25 level by 95% in experimental rats compared to the control. The immunoexpression of Hsp70 found no qualitative and quantitative differences in control and experimental lumbar spinal cords. Taken together our results show that (1) rat motoneurons survived after 35-day antiorthostatic hind limb suspension and the changes in neurons had a mostly functional character, and (2) the increased immunoexpression of Hsp25 can be considered as the anti-apoptotic factor. 相似文献

Mouse calvarial cells grown under simulated microgravity conditions (neutral buoyancy) show preferential differentiation towards the osteoclast lineage, as defined by surrogate mRNAs, bone nodule growth and TRAP+ cells, when compared with cells cultured under normal gravity conditions. This effect was suppressed in cultures which contained the immunoregulatory molecule CD200, and conversely enhanced by anti-CD200 mAb. Concomitant increases occur in expression of inflammatory cytokines, and their mRNAs, under simulated microgravity conditions. Again cultures containing exogenous CD200 showed suppressed cytokine and cytokine mRNA expression. Further alterations in osteoclastogenesis were seen using cells isolated from cytokine-receptor knockout mice. We conclude that, as assessed by altered expression of mRNAs associated with osteoblast differentiation, CD200:CD200R interactions play an important regulatory role in the enhanced osteoclastogenesis seen under simulated microgravity conditions, with changes in cytokine expression further modulating this effect. 相似文献

The descent in the atmosphere belongs to the most complicated phase of flight and presents stringent requirements for spaceplane systems. The enhanced requirements for reliability, the necessity of aircraft-type landing and a number of other reasons lead to the necessity of maximum involvement of the crew by means of creating, along with fully automatic, manual and director control systems and providing the crew with the possibility to monitor the systems operation and to actively override them. Participation of man in performance of such tasks during the atmospheric descent is hampered by a number of major difficulties caused by alteration of his state under the g-load effect.To evaluate the utmost capabilities of the manual and director control systems and to make rational selection of indication systems and controls it is necessary to create special flight simulation complexes comprising a hybrid computer complex capable of simulating vehicle trajectory and angular motions and the computer complex controlled centrifuge equipped with a crew-cabin, which would allow real-time simulation of the g-load effect upon the crew.This paper describes the structure of such complex, the distribution of tasks between the analog and digital computers and presents some results from evaluation of manual and director control system characteristics. 相似文献

An integrated analysis of both airborne and field short-wave infrared hyperspectral measurements was used in conjunction with conventional field mapping techniques to map hydrothermal alteration in the central portion of the Mount Painter Inlier in the Flinders Ranges, South Australia. The airborne hyperspectral data show the spatial distribution of spectrally distinct minerals occurring as primary minerals and as weathering and alteration products. Field spectral measurements, taken with a portable infrared mineral analyzer spectrometer and supported by thin-section analyses, were used to verify the mineral maps and enhance the level of information obtainable from the airborne data. Hydrothermal alteration zones were identified and mapped separately from the background weathering signals. A main zone of alteration, coinciding with the Paralana Fault zone, was recognized, and found to contain kaolinite, muscovite, biotite, and K-feldspar. A small spectral variation associated with a ring-like feature around Mount Painter was tentatively determined to be halloysite and interpreted to represent a separate hydrothermal fluid and fluid source, and probably a separate system. The older parts of the alteration system are tentatively dated as Permo-Carboniferous. The remote sensing of alteration at Mount Painter confirms that hyperspectral imaging techniques can produce accurate mineralogical maps with significant details that can be used to identify and map hydrothermal activity. Application of hyperspectral surveys such as that conducted at Mount Painter would be likely to provide similar detail about putative hydrothermal deposits on Mars. 相似文献

According to the flight characteristics of vehicle under high dynamic conditions, the impact of linear, angular, and vibrational motion on star sensor imaging was researched. Then, the mathematical model of motion blur corresponding to various flight conditions was deduced, and the star centroiding accuracy affected by motion blur was analyzed subsequently. On this basis, the mechanism of multiple blur was discussed. In order to ensure the accuracy of star sensor in high dynamic environment, a restoration method was proposed to remove the multiple blur. With the vast experience gained by Aerospace Community in the last five decades, the natural future course of action will be to expand Space Exploration. Our understanding of Moon is relatively better with a number of unmanned satellite missions carried out by the leading Space Agencies and manned missions to Moon by USA. Also a number of unmanned satellite missions and surface rover missions were carried out to Mars by those Space agencies generating many new details about Mars. While the future exploration efforts by global community will also be centered obviously on Moon and Mars, it is noteworthy that already NASA had declared its plans for establishing a Surface Base on Moon and developing the technical infrastructure required. Surface Bases on Moon and Mars give rise to a number of strategic, technical and ethical issues both in the process of development, and in the process of establishing the bases. The strategic issues related to Moon and Mars Surface Bases will be centered around development of enabling technologies, cost of the missions, and international cooperation. The obvious path for tackling both the technological development and cost issues will be through innovative and new means of international cooperation. International cooperation can take many forms like—all capable players joining a leader, or sharing of tasks at system level, or all players having their independent programmes with agreed common interfaces of the items being taken to and left on the surface of Moon/Mars. Each model has its own unique features. Among the technical issues, the first one is that of the Mission Objectives—why Surface Bases have to be developed and what will be the activity of crew on Surface Bases? Surface Bases have to meet mainly the issues on long term survivability of humans on the Mars/Moon with their specific atmosphere, gravity and surface characteristics. Moon offers excellent advantages for astronomy while posing difficulties with respect to solar power utilization and extreme temperature variations. Hence the technical challenges depend on a number of factors starting from mission requirements. Obviously the most important technical challenge to be addressed will be in the areas of crew safety, crew survivability, adequate provision to overcome contingencies, and in-situ resource utilization. Towards this, new innovations will be developed in areas such as specialized space suits, rovers, power and communication systems, and ascent and descent modules. The biggest ethical issue is whether humankind from Earth is targeting ‘habitation’ or ‘colonization’ of Moon/Mars. The next question will be whether the in-situ resource exploitation will be only for carrying out further missions to other planets from Moon/Mars or for utilization on Earth. The third ethical issue will be the long term impact of pollution on Moon/Mars due to technologies employed for power generation and other logistics on Surfaces. The paper elaborates the views of the authors on the strategic, technical and ethical aspects of establishing Surface Bases and colonies on Moon and Mars. The underlying assumptions and gray areas under each aspect will be explained with the resulting long-term implications. 相似文献
