二氯荧光素二聚体测定蛋白质

研究了阴离子染料二氯荧光素在阳离子表面活性剂十六烷基三甲基溴化铵（CTAB）存在时溶液的吸收光谱和荧光光谱特性。结果表明，二氯荧光素及其自聚的二聚体可平衡转化，在表面活性剂的存在下，牛血清白蛋白（BSA）的定量加入可调节这一平衡，从而引起体系荧光有规律的变化。还考查了二氯荧光素自聚体系测定蛋白质的影响因素和最佳实验条件，建立了测定牛血清白蛋白的新方法。在最佳实验条件下，对BSA测定的线性范围是0-25mg／L，检出限0．05mg／L。该方法灵敏度高、选择性好、线性范围较宽。

Measurements of Protein with Dimer of Dichlorofluorescein

The absorption spectrum and fluorescence spectrum of dichlorofluorescein in the solution of cationic surfactant cetyhrimethyl ammonium bromide （CTAB） were studied. The results showed that there is a self-polymerization equilibrium system between the anion of the fluorescent dye-dichlorofluorescein and its dimer. In the presence of a cationic surfactant, for example CTAB, the above mentioned equilibrium could be adjusted by the quantitative addition of protein, a regular change in fluorescence intensity of the system was observed and the effects of interference substances and optimization of procedure were investigated. Thus, a new method for the determination of the protein was established. Upon optimal conditions, the linear range reaches 0-25 mg/L and the detection limit 0.05 mg/L. The significant features of this method are its high sensitivity, selectivity and the wide range of linearity.
