长期空间飞行过程中航天员处于狭小密闭的失重环境中,密闭环境对机体骨代谢与糖脂代谢的影响尚不明确.通过4人180天受控生态生保系统集成实验,分析长期处于密闭环境的4名志愿者血清中骨代谢、糖脂代谢指标变化,并根据骨与能量代谢相互调控理论,分析二者相关性.实验结果表明,长期密闭舱内生活影响了骨代谢与糖脂代谢,表现为骨形成指标(BGP,PICP,BAP)的下降趋势.入舱前中后期Insulin的变化幅度较大.脂代谢类指标也有下降趋势.相关性分析表明BGP与FRUC呈极显著的正相关(r=0.525,p=0.001);BGP与CHOL和LDL也呈显著正相关(r=0.376,p=0.024;r=0.391,p=0.018).长期密闭环境影响机体的骨代谢与糖脂代谢,且二者存在一定关联.   相似文献

通过对大鼠尾吊模型进行改进,研制出一种新型可调节体位的大鼠后肢去负荷悬吊装置,研究模拟微重力效应下体液分布变化对大鼠骨代谢的影响.将36只SD大鼠均分为对照组(CON)、头低位后肢去负荷组(HDT)、水平位后肢去负荷组(HH)和头高位后肢去负荷组(HUT)4组,实验21天后,利用DXA检测大鼠的骨密度(BMD).模拟微重力效应下的三组大鼠后肢均发生严重骨丢失,其中HH和HUT组后肢BMD显著大于HDT组.实验结果表明,体液分布变化可能在模拟微重力效应导致的骨丢失中起到重要作用,新型大鼠后肢去负荷悬吊装置能够调节大鼠体位(体液)进行模拟微重力效应研究.   相似文献

The program of the 7-day flight of the biosatellite Cosmos-1667 launched in July 1985 included experiments on two rhesus monkeys, ten Wistar SPF rats, ten newts, Drosophila flies, maize seedlings, lettuce sprouts, and unicellular organisms - Tetrahymena. The primate study demonstrated that transition to orbital flight was accompanied by a greater excitability of the vestibular apparatus and an increased linear blood flow velocity in the common carotid artery. The rat studies showed that atrophy of antigravity muscles and osteoporosis of limb bones developed even during short-term exposure to microgravity. The experiments on other living systems revealed no microgravity effects on the cell division rate, proliferative activity of cells of regenerating tissues and organs, energy metabolism of developing insects, structure or chemical composition of higher plant seedlings.  相似文献

The bone loss induced by microgravity is partly due to the decrease of mature osteoblasts. In the present study, we employed the random positioning machine (RPM) to simulate microgravity and investigated the acute effects of simulated microgravity on the differentiation of 2T3 preosteoblasts. Following 7 days’ culture under normal (1 g) condition, cells were exposed to simulated microgravity for 24 h. The results showed that 24 h treatment of simulated microgravity significantly decreased alkaline phosphatase (ALP) activity without changing the cell morphology. In addition, the mRNA expressions of osteogenic genes, including runt-related gene 2 (Runx2), osterix, osteocalcin (OC), type I collagen (Col I) and bone morphogenetic protein (BMP), were dramatically downregulated. Moreover, western blot analysis of total extracellular signal-regulated kinase (Erk) and phosphorylated Erk (p-Erk) indicated that p-Erk level, which represents the Erk activation status, was increased. Taken together, our results suggested that acute exposure to simulated microgravity inhibited osteoblast differentiation through modulating the expression of osteogenic genes and the Erk activity. These findings provide new insight for bone loss due to microgravity and unloading.  相似文献

The effect of hypergravity on the white blood cell (WBC) line of mice was investigated by use of horizontal centrifuge. Several sets of experiments were performed, in which the parameters measured were the WBC and differential cell count in the peripheral blood. In another experiment, lymphocyte counts from the spleen, lymph nodes, and the thymus were measured. The needed samples were taken from the mice during a stay of 7-40 days under a hypergravity of 1.6G. The test groups that were placed on the arms of the centrifuge (1.6G) were compared with stationary control groups (1G) and a rotating control group located at the center of the centrifuge (1G). Such a comparison revealed the test animals to be deficient on all counts, to wit, showing a decrease in total number of WBC's, a decrease in lymphocyte number in the peripheral blood and a decrease in the number of lymphocyte in the spleen and thymus. The decrease of lymphocytes in peripheral blood was characterized by two different slopes--an early and temporary decrease at the first days of the experiment evident in both test and rotating control groups followed by a temporary increase, and a later persistent decrease, evident only in the test group, while in the rotating control lymphocyte counts reverted to normal. There were no significant differences in monocyte or neutrophil counts, except for a temporary increase in the number of neutrophils which peaked on the seventh day. In order to evaluate the effect of hypergravity on restoration of hematopoiesis following hematopoietic suppression, 5-fluoro-uracil (5-FU) was administered i.v. to both the experimental and control mice. Suppression of bone marrow was observed in all groups injected with 5-FU, but while there was later an increase in cell counts in the control groups, there was no such increase in the test group subjected to hypergravity.  相似文献

The rapid loss of bone mass is one of the serious problems which have to be solved before long-lasting manned spaceflights may be considered. In this paper we describe investigations in which we have checked whether the bone loss in astronauts as well as in osteoporotic patients may be related to abnormalities in a recently discovered calcium-binding protein, named osteocalcin. It was observed that in all subjects of a limited number of osteoporotic patients, the amount of calcium-binding groups (Gla-residues) in the circulating osteocalcin was substantially reduced. The Gla-content could be normalized, however, by the oral administration of vitamin K (1 mg/day). We also analyzed the Gla-content of plasma-osteocalcin from 4 astronauts before and after the D-1 mission. The amount of Gla-residues was reduced by more than 50% in the post-flight samples. It seems probable, that an increased vitamin K-intake by the astronauts will correct the observed abnormality, but whether this will lead to a decrease of the microgravity-induced bone-loss remains to be seen.  相似文献

Statistical analyses of the influence of Solar and geomagnetic activity, sector structure of the interplanetary magnetic field and galactic cosmic ray Forbush effects on car accident events in Poland for the period of 1990-1999 have been carried out. Using auto-correlation, cross-correlation, spectral analyses and superposition epochs methods it has been shown that there are separate periods when car accident events have direct correlation with Ap index of the geomagnetic activity, sector structure of the interplanetary magnetic field and Forbush decreases of galactic cosmic rays. Nevertheless, the single-valued direct correlation is not possible to reveal for the whole period of 1990-1999. Periodicity of 7 days and its second harmonic (3.5 days) has been reliably revealed in the car accident events data in Poland for the each year of the period 1990-1999. It is shown that the maximum car accident events take place in Poland on Friday and practically does not depend on the level of solar and geomagnetic activities.  相似文献

The purpose of this study is to reveal the effect of chronic hypergravity exposure on the bone formation and the bone metabolism when mammals produce offspring in a 2 G environment. We measured the length and width of the thighbone, the length of the pelvis, the width of the pelvic cavity and the width of the fourth cervical vertebra on the second (F2) and the third (F3) generation mice bred in a 2 G environment every ten days from 20 days old to 60 days old in an experiment on bone formation. In an experiment on bone metabolism, we measured calcium and phosphorus in the bones of the F3 in the 2 G group. Ratios of the thighbone length, pelvis length, pelvic cavity width, and fourth cervical vertebra width versus the body length were calculated. These ratios were higher in the 2 G group than the control group during all measuring periods. Calcium and phosphorus concentrations in the thighbone and the lumbar vertebra were lower in the 2 G group than in the control group. However, the calcium and phosphorus concentrations in the cervical vertebrae of the 2G group were higher. These results suggest that the influence of gravity load may vary in the bones.  相似文献

The effects of gravity on bone metabolism are unclear, and little has been reported about the effects of hypergravity on the mature skeleton. Since low gravity has been shown to decrease bone volume, we hypothesized that hypergravity increases bone volume. To clarify this hypothesis, adult female rats were ovariectomized and exposed to hypergravity (2.9G) using a centrifugation system. The rats were killed 28 days after the start of loading, and the distal femoral metaphysis of the rats was studied. Bone architecture was assessed by micro-computed tomography (micro-CT) and bone mineral density was measured using peripheral quantitative CT (pQCT). Hypergravity increased the trabecular bone volume of ovariectomized rats. Histomorphometric analyses revealed that hypergravity suppressed both bone formation and resorption and increased bone volume in ovariectomized rats.  相似文献

Animal models are frequently used to assist in the determination of the long- and short-term effects of space flight. The space environment, including microgravity, can impact many physiological and immunological system parameters. It has been found that ground based models of microgravity produce changes in white blood cell counts, which negatively affects immunologic function. As part of the Center of Acute Radiation Research (CARR), we compared the acute effects on white blood cell parameters induced by the more traditionally used animal model of hindlimb unloading (HU) with a recently developed reduced weightbearing analog known as partial weight suspension (PWS). Female ICR mice were either hindlimb unloaded or placed in the PWS system at 16% quadrupedal weightbearing for 4 h, 1, 2, 7 or 10 days, at which point complete blood counts were obtained. Control animals (jacketed and non-jacketed) were exposed to identical conditions without reduced weightbearing. Results indicate that significant changes in total white blood cell (WBC), neutrophil, lymphocyte, monocyte and eosinophil counts were observed within the first 2 days of exposure to each system. These differences in blood cell counts normalized by day 7 in both systems. The results of these studies indicate that there are some statistically significant changes observed in the blood cell counts for animals exposed to both the PWS and HU simulated microgravity systems.  相似文献

Data of galactic cosmic rays, solar and geomagnetic activities and solar wind parameters on the one side and car accident events (CAE) in Poland on the other have been analyzed in order to reveal the statistical relationships among them for the period of 1990-2001. Cross correlation and cross spectrum of the galactic cosmic ray intensity, the solar wind (SW) velocity, Kp index of geomagnetic activity and CAE in Poland have been carried out. It is shown that in some epochs of the above-mentioned period there is found a reliable relationship between CAE and solar and geomagnetic activities parameters in the range of the different periodicities, especially, 7 days. The periodicity of 7 days revealed in the data of the CAE has the maximum on Friday without any exception for the minimum and maximum epochs of solar activity. However, the periodicity of 7 days is reliably revealed in other parameters characterizing galactic cosmic rays, SW, solar and geomagnetic activities, especially for the minimum epoch of solar activity. The periodicity of 3.5 days found in the series of CAE data more or less can be completely ascribed to the social effects, while the periodicity of 7 days can be ascribed to the social effect or/to the processes on the Sun, in the interplanetary space and in the Earth's magnetosphere and atmosphere.  相似文献
